Embodiment 1

[0046] A method for rapidly propagating through tissue culture of kaempferia florets, comprising the steps of:

[0047] a) Pre-treatment of explants: collect the rhizomes of Kaempferia florida and clean them with tap water. After cutting off the fibrous roots on the rhizomes, put them into carbendazim powder solution with a mass concentration of 0.3% for 30 minutes and then put them into 0.3% carbendazim powder solution for disinfection. % potassium permanganate solution for 30 minutes, take out and dry the surface moisture, such as figure 1 shown. Then put it into the washed and disinfected river sand, and germinate at a constant temperature of 30°C in an incubator, such as figure 2 shown. Wash the rhizome buds, and take the base of the buds as the explant material.

[0048] For the washed and disinfected river sand described in step a), the river sand is first rinsed with sterilized water for 3 times, then soaked and disinfected with 0.1% potassium permanganate solution ...

Embodiment 2

[0060] A method for rapidly propagating through tissue culture of kaempferia florets, comprising the steps of:

[0061] (a) Pre-treatment of explants: collect the rhizomes of Kaempferia florida and clean them with tap water, cut off the fibrous roots on the rhizomes, put them in 0.1% carbendazim powder solution for immersion and disinfection for 40 minutes, and then put them in Soak and disinfect in 0.1% potassium permanganate solution for 40 minutes, remove and dry the surface moisture. Then put it into the washed and disinfected river sand, and germinate at a constant temperature of 32°C in an incubator. Wash the rhizome buds, and take the base of the buds as the explant material.

[0062] The washed and disinfected river sand described in step (a) is to wash the river sand with sterilized water for 4 times, then soak and disinfect the river sand with 0.3% potassium permanganate solution for 3 hours, and then dry it at room temperature.

[0063] (b) bud base proliferation:...

Embodiment 3

[0074] A method for rapidly propagating through tissue culture of kaempferia florets, comprising the steps of:

[0075] (a) Pre-treatment of explants: collect the rhizomes of Kaempferia florida and clean them with tap water, cut off the fibrous roots on the rhizomes, put them in 0.2% carbendazim powder solution for immersion and disinfection for 30 minutes, and then put them in a mass concentration Soak and disinfect in 0.2% potassium permanganate solution for 30 minutes, remove and dry the surface moisture. Then put it into the washed and disinfected river sand, and germinate at a constant temperature of 33°C in an incubator. Wash the rhizome buds, and take the base of the buds as the explant material.

[0076] The washed and disinfected river sand described in step (a) is to wash the river sand with sterilized water for 4 times, then soak and disinfect the river sand with 0.2% potassium permanganate solution for 4 hours, and then dry it at room temperature.

Abstract

The invention discloses a tissue culture and rapid propagation method of flowerlet hedychium coronarium. The method comprises the following steps: pre-treating an explant, proliferating a bud base, inducing cluster buds, carrying out subculture proliferation, carrying out rooting and culturing sound seedlings, and exercising the seedlings and transplanting. The method comprises the specific steps:inoculating a disinfected bud base explant into a proliferation culture medium and culturing until a spherical shape is formed on the base of the explant; cutting the proliferated bud base and transferring the proliferated bud base to a cluster bud induced culture medium to induce the cluster buds; when subculture cluster bud seedlings are 4cm to 6cm high, cutting off the cluster buds and transferring the cluster buds to a sound seedling rooting culture medium and culturing; when the seedlings on the sound seedling rooting culture medium are 6cm to 8cm high, exercising the seedlings in a greenhouse under natural illumination for 7d to 10d; taking out the seedlings and washing the culture medium on roots; culturing to obtain transplanted seedlings. According to the tissue culture and rapidpropagation method disclosed by the invention, tissue culture seedlings capable of keeping good properties of stock plants can be obtained in a short period; the tissue culture and rapid propagationmethod has small damages to the stock plants and is low in investment and high in yield; a lot of flowerlet hedychium coronarium tissue culture seedlings can be provided.

Description

technical field [0001] The invention relates to small flower kaempferia, in particular to a tissue culture rapid propagation method of small flower kaempferia, which belongs to the technical field of plant tissue culture rapid propagation. Background technique [0002] Zingiberaceae plants can be propagated by sowing, cutting rhizomes, cuttings and tissue culture (Gao Jiangyun et al., 2006; Xiong Youhua et al., 2005, 2007; Wu Fanhua et al., 2009; Huang Sai et al., 2016; Wu Delin et al., 2016 ; Li Dongmei et al., 2017). Kaempferia parviflora belongs to the genus Kaempferia (Kaempferia) of Zingiberaceae, with a plant height of about 30cm, clustered leaves, oval leaves, green leaf surface, red leaf margin, and dark red markings on the back of the leaf; In the bract, most of the flowers are small, open in sequence, the flowers are white, and the lips have purple-red markings; the flowering period is May-October; the rhizomes are massive, medicinal; foliage, good plant shape, be...
