Embodiment Construction

[0027]In order to make the objectives, technical solutions and advantages of the present invention clearer, the following further describes the present invention in detail with reference to the accompanying drawings and embodiments.

[0028]Such asfigure 1 As shown, the method for tracking the movement of vesicles in living cells sequentially includes two steps: (1) vesicle identification and (2) trajectory connection;

[0029]The (1) vesicle recognition includes four steps in sequence: (1.1) wavelet transform extraction, (1.2) watershed segmentation, (1.3) centroid extraction, and (1.4) noise removal;

[0030]The (2) trajectory connection includes two steps (2.1) position prediction and (2.2) local linear optimization in turn.

[0031]By labeling vesicles with fluorescent proteins, a series of dynamic images of vesicle movement can be collected with the help of a fluorescent microscope. An example image is shown in Figure 2(a), and the original image is denoted as A0.

Abstract

The invention belongs to the technical field of biology, and particularly relates to a method for tracking movements of vesicae in living cells. The method comprises a vesica recognition step (1) anda track connection step (2) in sequence, wherein the vesica recognition step (1) comprises a wavelet transform extraction step (1.1), a watershed segmentation step (1.2), a centroid extraction step (1.3) and a miscellaneous point removal step (1.4) in sequence; and the track connection step (2) comprises a position prediction step (2.1) and a local linear optimization step (2.2) in sequence. According to the method for tracking movements of vesicae in living cells, the movement tracks of a large number of vesicae are connected; and by adoption of the method, the movement tracks of the vesicaein living cells are analyzed, so that the law of life activities is disclosed and data support is provided for screening diabetes-related drugs.

Description

Technical field[0001]The invention belongs to the field of biotechnology, and specifically relates to a method for tracking the movement of vesicles in living cells.Background technique[0002]Life lies in movement, and life is composed of cells, so the vitality of cells determines the vitality of life. Vesicles are responsible for the transport of substances in cells, so abnormalities in the transport mechanism of vesicles can lead to diseases, such as metabolic diseases such as diabetes. Therefore, by tracking the trajectory of vesicles in living cells, on the one hand, we can have a deeper understanding of the laws of life activities, on the other hand, it can also provide analytical data for the screening of diabetes-related drugs. Due to the large number of vesicles in the cell and the complicated movement, it is always difficult to track the vesicles accurately and quickly.Summary of the invention[0003]In view of the above technical problems, the present invention proposes a met...
