Embodiment 1
[0166] This example demonstrates the analysis of microbiota from healthy versus diseased subjects to identify a significantly increased prevalence of A. hadrus in healthy subjects compared to subjects with IBD.

[0168] Research Design and Sampling
[0169] The study and sample collection were approved by the Clinical Research Ethics Committee of University College Cork Teaching Hospitals. All participating individuals provided informed consent before gastrointestinal examinations were performed at Cork University Hospital or Bon Secours Hospital in Cork, Ireland. Diagnosis of CD or UC and inflammatory status is based on macroscopic examination of the GI tract during standard clinical colonoscopy. Adults (>=18 years old) who were routinely monitored and not diagnosed with any digestive disease or syndrome were considered as healthy controls. Biopsy samples were obtained from inflamed tissue from adult IBD patients or healthy tissue from control subjects...
Embodiment 2
[0194] Use of A. hadrus for treating inflammatory bowel disease
[0195] In this example, the glucose tolerance test (GTT) was performed on a high-fat diet-induced obese mouse model to study the effect of administering live bacteria on glucose homeostasis. Mice were prepared and used as described in WO2019067087 (which is incorporated herein by reference in its entirety).
[0196] IBD was induced in mice by supplementing drinking water with 3% sodium dextran sulfate for 7 days. For each daily treatment, A. hadrus liquid cultures were grown to an optical density of 0.4–0.5 and then pelleted by centrifugation. Bacteria were resuspended in phosphate-buffered saline and mice were administered 100 μl by oral gavage starting on day 8. Mice were treated daily for 1 week with A. hadrus suspension or phosphate-buffered saline alone as negative controls. Five days after bacterial treatment, live mice were scored for colitis using endoscopy. Endoscopic injury scores were determined b...
