The preparation method comprises the following steps of culturing vero cells; a virus culture step: adding the cell culture fluid of the porcine epidemic diarrhea virus variant shxx1902 into the vero cell culture fluid, carrying out dilution passage, and culturing for 72 hours for later use; an immunogen preparation step: adding 0.2% formaldehyde into the virus liquid, and inactivating the virus at 37 DEG C; then adding into a 96-well plate paved with vero cells, and culturing in a CO2 incubator; collecting a cell supernatant, fixing the cells by using cold acetone, adding the collected supernatant into a 96-well plate paved with vero cells, and continuously culturing; and removing the supernatant, fixing the cells with cold acetone, adding an alumina gel adjuvant after inactivation verification is qualified, and mixing for use.
