specific Embodiment approach 1

[0023] Embodiment 1: The method for analyzing and testing the content of somatic cells in raw milk in this embodiment is carried out according to the following steps:

[0024] (1) Preparation of reaction reagents: respectively prepare a buffer solution with a pH value of 7.5 to 10 and a concentration of 0.01 to 1M, an ester solution with a concentration of 1 to 100mM, and a diazonium salt solution with a concentration of 1 to 100mM, wherein the buffer solution is mixed with a concentration of 0.01-30mM reaction aid;

[0025] (2) Reaction: Add the raw milk sample into the reaction cup, and then add the reaction reagents prepared in step (1) into the reaction cup in turn, that is, the buffer solution, the ester solution, and the diazonium salt solution. The volume percentage of the raw milk sample is 20~ 80%, the adding volume of buffer solution, ester solution and diazonium salt solution can be determined according to the concentration of the prepared solution, the final concen...

specific Embodiment approach 2

[0030] Embodiment 2: The filter paper can be soaked in the reaction reagent prepared in the first step (1) of Embodiment 1 for 10-30 minutes, and then heated and dried on tin foil after completely soaking to make reaction test paper, which is stored dry.

[0031] The method of using the reaction test paper is to drip the raw milk sample onto the test paper, or immerse the test paper in the sample for 1-3 seconds, then lay the test paper flat, visually observe or use an instrument to collect the color change information of the test paper during the reaction, and use Standard microscope method or its equivalent method, make a standard color comparison chart by visual inspection, or establish a fixed functional relationship between the color change information and the number of somatic cells in the reaction process by machine measurement, as the detection standard; mix the raw milk to be tested with the test paper Perform the reaction, visually observe or use an instrument to coll...

Embodiment approach

[0035] Preferred embodiment: the ester solution is one of pyrrolyl ester, indoxyl ester or polynaphthyl acetate. Other steps and parameters are the same as those in Embodiment 1 or Embodiment 2.

[0036] Preferred embodiment: the concentration of the ester solution in the reaction reagent is 10-80 mM, and other steps and parameters are the same as those in Embodiment 1 or Embodiment 2.

[0037] Preferred embodiment: the diazonium salt is 1,2-diazooxynaphthalene-4-sulfonic acid or 2,4-dichloroaniline diazonium salt. Other steps and parameters are the same as those in Embodiment 1 or Embodiment 2.

[0038] A preferred embodiment: the concentration of the diazonium salt in the reaction reagent is 10-80 mM, and other steps and parameters are the same as those in Embodiment 1 or Embodiment 2.

[0039] Preferred embodiment: the buffer is: one of borate buffer, boric acid-sodium hydroxide buffer, tris-HCl buffer or phosphate buffer. Other steps and parameters are the same as those...

Abstract

The invention discloses a method for analyzing and testing body cell content in raw milk. The method comprises the steps of preparing a reaction reagent, reacting, establishing a standard and detecting the raw milk to be detected. Besides a detecting method with a solution reagent, the invention also provides a method for detecting with reaction test paper prepared by the reaction reagent. The method disclosed by the invention has the advantages that: detection speed is fast, and the number of body cells in the raw milk can be quantitatively determined within one minute most quickly, thus being applicable to online detection; and detection cost is low, detection result is accurate, environmental requirement is low, and detection operation is simple and convenient.

Description

technical field [0001] The invention relates to the field of biochemical detection, in particular to an analysis and test method for somatic cell content in raw milk. Background technique [0002] Somatic cell count in raw milk refers to the total number of cells per milliliter of raw milk. The somatic cell count is an important indicator to measure the quality of raw milk and the health of the mammary glands of the animals that supply raw milk. A simple, low-cost and high-accuracy detection method is needed. At present, there are many detection methods for somatic cell count or mastitis in raw milk used in the dairy industry at home and abroad. Common methods include microscope detection (standard microscope method and fluorescence microscope method), electronic counter detection method, California cell count method (CMT), Wisconsin Mastitis Test (WMT); the standard microscope method is the benchmark method for the detection of somatic cell count in my country. This method ...
