Abstract

The invention discloses a method for obtaining callus and suspension cells under induction of Taxus chinensis test tube plantlet. The method comprises the following steps: (1) seed testa peeling: mechanical cracking is utilized and the testa is peeled manually; (2) explants disinfection: disinfection is carried out for 90 seconds with 70% alcohol and carried out for 12 minutes with 0.1% mercuric chloride, and washing is carried out by 5 times with sterile water for standby; (3) pretreatment of seed: seed soaking is carried out by 1-5 days at normal temperature hermetically with sterile water; (4) zygotic embryo separation; (5) induction of test tube plantlet: embryo is inoculated to a test tube plantlet induced culture substrate; (6) induction of callus; (7) subculture of callus; and (8) suspension culture of callus.
