The method comprises the following steps of: inoculating schizophyllum commune on a PDA (potato dextrose agar) plating medium to perform rejuvenation and activation cultivation to obtain a plating strain; inoculating the plating strain to a seed medium, and culturing to obtain a primary strain; inoculating the primary strain in the seed medium, and culturing to obtain a secondary strain; inoculating the secondary strain on a basic medium and fermenting to obtain fermented mash; and extracting and purifying the fermented mash to obtain the compound polysaccharide, wherein the seed medium comprises the following components in 1 kilogram of seed medium: 50g of glucose, 5g of monopotassium phosphate, 2.4g of magnesium sulfate, 3g of yeast extract, and the balance of tap water; and the asic medium comprises the following components in 1 kilogram of basic medium: 50g of glucose, 5g of monopotassium phosphate, 2.4g of magnesium sulfate, 4.5g of beef extract, 3g of calcium carbonate, 5g of oat bran and the balance of tap water.
