The method for measuring the diffusion coefficient of the dissolved oxygen by utilizing the device comprises the following steps: placing a to-be-measured biological membrane sample into the biological membrane measuring chamber, and introducing nitrogen to reduce the concentration of oxygen in the system to be the minimum; then changing intake gas into oxygen, and meanwhile using a dissolved oxygen microelectrode to record the curve about time-dependent change of the dissolved oxygen in the biological membrane; repeating the test in different positions and different depths of the biological membrane, so as to obtain multiple pieces of sample data; simulating the process through a mathematical model and fitting model data and data measured in the test, and calculating the diffusion coefficient of the dissolved oxygen.
