Embodiment Construction

[0024] The invention utilizes the influenza virus VLPs platform to fuse the main structural protein G of carp spring viremia virus with a part of influenza virus HA to form a new fusion protein HA-G, and then simultaneously express HA-G and M1 proteins in insect cells , automatically assembled into VLPs. Using VLPs expressing fusion protein HA-G as antigen, a novel genetically engineered vaccine against carp spring viremia virus was prepared. The novel genetic engineering vaccine of carp spring viremia virus may also include an adjuvant.

[0025] Based on the Bac-to-Bac insect baculovirus expression system, the invention constructs the fusion protein HA-G from the influenza virus HA protein and carp spring viremia virus G protein, and expresses the influenza virus matrix protein M1 and the fusion protein HA at the same time -G, the two co-assemble automatically to construct VLP empty shell particles similar to influenza virus.

Abstract

The invention relates to SVCV (Spring viraemia of carp) resisting VLPs (virus-like particles), a vaccine and a preparation method thereof. The SVCV resisting VLPs are composed of influenza virus matrix protein M1 and fusion protein HA-G which are expressed at the same time; the fusion protein HA-G is built through influenza virus HA protein and SVCV G protein, and the nucleotide sequence of the fusion protein HA-G is shown in SEQ ID NO.3, 5, 7, 9, 11 or 13. The preparation method of the SVCV resisting VLPs comprises the following steps: preparing a fusion gene HA-G; preparing a recombinant baculovirus shuttle vector rBacmid; preparing recombinant insect baculovirus; obtaining VLPs which can express the influenza virus matrix protein M1 and the fusion protein HA-G simultaneously. The vaccine comprises any one virus-like particle. An organism immunity system can cause strong response by blending VLPs and the vaccine, the immunity is strong, the time of duration is long, the SVCV can be prevented, and high safety is achieved.

Description

technical field [0001] The invention relates to the field of virus-like particles, in particular to a virus-like particle against carp spring viremia virus, a preparation method and a vaccine thereof. Background technique [0002] Spring viraemia of carp (SVC) is also known as swim bladder inflammation (SBI), also known as acute infectious ascites (acute infectious drop), carp infectious ascites (infectious drop of carp, IDC) ), carp infectious ascites (ascites infectious cyprinorum), hemorrhagic sepicemia, infectious ascites or spring virus disease, etc. The disease is an acute hemorrhagic infectious viral disease. Often endemic in carps, especially carps, the disease usually occurs in spring outbreaks and causes mortality in juveniles and adults. The disease is caused by a rhabdovirus, carp spring viremia virus (SVCV for short). The virus has an envelope, the virus size is 180×70nm, contains single-stranded RNA, and the floating density in CsCl is 1.195-1.200g / mL. Acco...
