Problems solved by technology

[0007] Aiming at the lack of high-quality tomato varieties with TYLCV resistance on the market, it is difficult to aggregate the high-pigment gene dg and the disease-resistant gene Ty by using traditional techniques, and at the same time remove the linkage burden of unfavorable agronomic traits

Embodiment

[0050] The principle and process of the invention are described in detail through the following examples.

[0051] (1) Identification method of yellow leaf curl virus (TYLCV) resistance

[0052] This program introduces a method for identification of tomato plants resistant, moderately resistant and susceptible to TYLCV.

[0053] Plants are grown in the field for natural inoculation by Bemisia tabaci. It is a preferred method to plant and inoculate local viruses in natural high-incidence fields to identify plant resistance. The disease symptoms of the plants after inoculation were observed in the field, and the disease index was counted. Symptoms of TYLCV viral disease are usually graded 0-4. Grade 0: no symptoms; Grade 1: slight yellowing of leaves; Grade 2: obvious yellowing of leaves with curling; Grade 3: growth retardation of plants, severe yellowing and curling of leaves; Grade 4: severe growth retardation of plants with Yellowish, curled leaflets.

Abstract

The invention discloses a method for breeding a novel tomato line rich in lycopene and resisting against a yellow leaf curl virus. The method comprises the following steps: screening a recessive mutant gene dg specificity Caps molecular marker according to tomato genome DNA (Deoxyribonucleic Acid) sequence signatures; screening a pair of Caps molecular markers which are closely linked with the anti-TYLCV (Tomato Yellow Leaf Curl Virus) gene Ty-3; hybridizing a tomato material containing the TYLCV resistance gene and the tomato mutant dg material with a common tomato good selfing line; conducting multi-generation selfing and taking the dg and Ty-3 molecular markers as basis to screen single plants; at last, obtaining the excellent selfing line aggregating the dg gene and the Ty-3 gene. The method can also be applied to a hybrid which is obtained by utilizing the selfing line to be hybridized with the other tomato selfing lines.

Description

technical field [0001] The invention belongs to the field of agricultural breeding, in particular to a tomato breeding method for simultaneous transfection of high lycopene gene and anti-yellow leaf curl virus gene by using molecular markers. Background technique [0002] Tomato is one of the most important crops with high economic value in the world, and it is one of the vegetable crops with the highest annual yield in the world. Many cultivars with excellent traits have been bred using traditional breeding methods, such as resistance to various diseases, stress resistance, storage and transportation, and nutrient-enhanced tomato varieties. In modern society, a trait of great interest is the development of secondary metabolites that can confer health benefits in humans. Tomatoes contain a variety of carotene with antioxidant effects, which have a strong health care effect on the human body. Epidemiological studies have shown that lycopene can reduce the efficacy of some c...
