The method comprises the steps of selecting the wooden box which is 80 cm in length, 60 cm in width and 40 cm in height, placing base materials into the wooden box, then placing the Northern Star No.2 red earthworms in the box for breeding, wherein the thickness of the base materials placed in the wooden box ranges from 30 cm to 35 cm, spraying water frequently during the breeding period so that the relative humidity of the base materials can be kept within the range from 60% to 70%, using straw matting and old jute bags for covering an upper end opening of the wooden box for shading, and controlling the temperature within the range from 23 DEG C to 25 DEG C.
