Embodiment 1

[0021] Embodiment 1: Add the fermentation of vitamin C

[0022] A method for utilizing vitamin C to improve Paenibacillus polymyxa to produce antifungal substances, comprising the steps of:

[0023] (1) Preparation of medium

[0024] Potato medium (PDA medium): Take 200g of peeled potatoes, cut into small pieces, add 1000ml of water to boil until it breaks when touched, collect the filtrate with 6 layers of gauze, add 20g of glucose, 20g of agar, dilute to 1000ml, divide Steam sterilization at 121°C for 20 minutes after loading;

[0025] Seed medium composition in g / L: starch 30, glucose 5, yeast extract powder 2, peptone 4, MgSO 4 . 7H 2 O0.5, NaCl0.5, K 2 HPO 4 1.5, steam sterilization at 121°C for 20 minutes;

[0026] Fermentation medium components in g / L: glucose 20, yeast extract powder 3, NaCl1, KH 2 PO 4 2, (NH4) 2 SO 4 1. MgSO 4 ·7H 2 O0.3, KCl0.2, CaCl 2 0.02, FeSO 4 ·7H 2 O0.002, MnSO 4 ·H 2 O0.002, pH 7.0, steam sterilization at 121°C for 20 minute...

Embodiment 2

[0036] Control group: Fermentation without vitamin C addition

[0037] (1), (2) and (4) are the same as embodiment 1 steps (1), (2) and (4);

[0038] (3) Put the first-grade seeds into a 250ml Erlenmeyer flask filled with 100mL fermentation medium at an inoculation density of 1% by volume, and ferment for 72 hours at a stirring speed of 200rpm and a temperature of 37°C. The biomass accumulation detected in 36h, 48h, 60h time period is as follows figure 1 As shown, the generated antifungal substances such as figure 2 shown.

[0039] in conclusion:

[0040] (1) Adding 10mg / L, 30mg / L, and 200mg / L vitamin C during the fermentation process promoted the biomass accumulation of P.polymyxaCJX-518;

[0041] (2) Adding 10mg / L, 30mg / L, and 200mg / L vitamin C during the fermentation process to promote the production of antifungal active substances by P.polymyxaCJX-518;

[0042] (3) Adding 30mg / L vitamin C has the best promotion effect. Compared with the control at 36h, 48h and 60h, t...

Abstract

The invention discloses a method for using an external source for promoting (Paenibacillus polymyxa) CJX-518 to produce antifungal matter by adding a reducing agent vitamin C, and belongs to the technical field of fermentation engineering. The method comprises the steps that in the fermentation process, (Paenibacillus polymyxa) CJX-518 with the preservation serial number of CGMCC NO.7096 is cultured in a seed culture medium, and first-grade seeds are obtained; the first-grade seeds are inoculated into a fermentation culture medium at the inoculation density of 1% by the volume percent, and fermentation is carried out for 72 h under the conditions that the stirring rotating speed is 200 rpm and the temperature is 37 DEG C; when fermentation is carried out for 4 h, different experimental groups are added with a vitamin C aqueous solution at the concentration of 10 mg / L, a vitamin C aqueous solution at the concentration of 30 mg / L and a vitamin C aqueous solution at the concentration of 200 mg / L respectively. By means of the method, accumulation of antifungal matter produced by P. polymyxa CJX-518 is promoted. Under the batched fermentation mode, the amount of the antifungal matter produced by P. polymyxa CJX-518 can be remarkably increased with the method.

Description

[0001] The invention is funded by Tianjin Natural Science Foundation 13JCYBJC25500. technical field [0002] The invention belongs to the technical field of fermentation engineering and relates to a method for improving the production of antifungal substances by Paenibacillus polymyxa by using vitamin C. Background technique [0003] Plant diseases caused by fungi have become the main obstacle to stable, high and high-quality yields of economic crops and medicinal plants. These will cause economic losses and produce mycotoxins that are harmful to the human body, making the prevention and treatment of fungal diseases a more important. Most of the currently used fungicides will cause environmental risks, so environmentally friendly biological control substances that can replace these chemicals have become a research hotspot. [0004] Fungal diseases are the most important type of plant diseases, accounting for more than 80% of plant diseases, about 30,000 species, resulting i...
