[0017] Current strategies to increase the accuracy of sequence information from single-molecule sequencing platforms are inadequate. In contrast, the multiple-pass sequencing (MPS) method disclosed in the present invention produces precise sequences while retaining the advantages of long read length and speed. MPS finds particular application utilizing nanopore sequencing and is also advantageously used in conjunction with other single-molecule sequencing methods such as nanochannel-based sequencing (e.g., Schmidt, 2004, "A nanoelectrode lined nanochannel for single molecule sequencing" (Thesis) card Negie Mellon University) or zero-mode waveguide sequencing (eg, SMRTPacific Biosciences). Thus, although the following discussion focuses on the application of MPS to nanopore sequencing, it should be understood that the same concatemer molecules, libraries and analysis methods can be used with any suitable SMS method.
