The method includes the steps that light field imaging and fluorescence imaging are conducted on a target sample by adopting a microscopic fluorescence imaging system to obtain the light field image and the fluorescence image, edge extraction and segmentation are conducted on a fluorescence image region of each detection target in the fluorescence image, and the fluorescence image region corresponding to each detection target in the fluorescence image is obtained; the cumulative gray value, the maximum gray value, and the average gray value of the fluorescence image region of each detection target, and the diameter value of the light field image region of each detection target are calculated, streaming clustering analyses are conducted based on the cumulative gray value, the maximum gray value, the average gray value and the light field diameter value.
