Embodiment 1

[0035] Embodiment 1: the screening 1 of feather degrading strain

[0036] Soil from a pile of sick and dead chickens was collected from a chicken farm in Pulandian.

[0037] The ABTNL-4 strain was obtained through enrichment culture, primary screening, secondary screening, and further isolation and purification.

[0038] Test rig and feather handling

[0039] Experimental device for crushing feathers: 220V, 3.6KW high-speed multifunctional grinder.

[0040] Feather treatment: The feathers collected at the live poultry slaughter point are rinsed with running water, dried at 55°C for 4-7 days, and set aside. Feathers are pulverized by a pulverizer and passed through a 100-mesh sieve for solid culture.

[0041] Preparation of enrichment medium: Add 5.0 g of beef extract, 10.0 g of peptone, 5.0 g of sodium chloride, and 1.0 g of dipotassium hydrogen phosphate into a conical flask containing 1000 ml of distilled water, and stir to dissolve on a magnetic stirrer. Then adjust the...

Embodiment 2

[0052] Embodiment 2: the screening 2 of feather degrading strain

[0053] Soil samples were collected from a pile of sick and dead chicken carcasses in a chicken farm in Pulandian.

[0054] The ABTNL-4 strain was obtained through enrichment culture, primary screening, secondary screening, and further isolation and purification.

[0055] Test rig and feather handling

[0058] Preparation of enrichment medium: Weigh 5.0g of beef extract, 10.0g of peptone, 5.0g of sodium chloride, and 1.0g of dipotassium hydrogen phosphate, add them into a conical flask containing 1000ml of distilled water, stir and dissolve on a magnetic stirr...

Embodiment 3

[0069] Embodiment 3: strain identification

[0070] The above-mentioned degradable feather strain identification process of the present invention is specifically as follows:

[0071] Identification of strains: use 16SrRNA method to identify microbial strains

[0072] Bacteria identification results: PCR amplification and sequencing: the strain was cultured to the exponential growth phase with broth medium as the substrate, the bacteria were collected by centrifugation, and the total DNA was obtained by boiling with a PCR machine. The primers used in the 16S rRNA PCR reaction system were universal Primers (27F: 5-AGAGTTTGATCCTGGCTCAG-3, 1492R: 5-GGTTACCTTGTTACGACTT-3,).

[0073] PCR reaction system

[0074]

[0075] PCR reaction program: pre-denaturation at 94°C for 5 min; denaturation at 94°C for 1 min, annealing at 56°C for 1 min, extension at 72°C for 2 min; cycle 29 in the second step; 10 min at 72°C, electrophoresis observation. The PCR products were sent to Shanghai...

Abstract

The invention provides a screening method for a feather degradation strain. The specific steps include: soil sample taking from a chicken farm site for accumulating chickens died of illness; enrichment; preliminary screening; secondary screening; separation and purification: observing the growth condition of microbial strains on a solid screening plate, selecting a dominant strain growing best, and selecting single colony for plate streaking; conducting streak culture to obtain the single colony, i.e. feather degradation strain ABTNL-4. The invention also provides an application method of the feather degradation strain, and the method includes: taking feather powder as the only carbon source and nitrogen source to prepare a fermentation medium, adding the feather degradation strain ABTNL-4 into the medium, and performing culture at 37DEG C, thus realizing feather degradation. The method provided by the invention can achieve a feather degradation rate up to 95%, compared with the physical-chemical process for feather degradation, the microbial method for feather degradation has the advantages of mild reaction conditions, little energy consumption, economical efficiency and environmental protection, at the same time avoids the loss of most amino acids, and improves the absorbability and utilizability feather degradation products.

Description

technical field [0001] The invention belongs to the technical field of microorganisms, and more specifically relates to a screening method and an application method of degradable feather strains. Background technique [0002] Feathers are keratinized derivatives of the epidermal cells of birds and birds, which make up 10% of their body weight. According to statistics, the annual slaughter volume of poultry in my country: 11.01 billion in 2010, 11.33 billion in 2011, 12.08 billion in 2012, 11.90 billion in 2013, 11.54 billion in 2014, and 14.352 billion in 2015. According to this trend, the feather production of poultry continues to increase. [0003] More than 90% of the main component of feathers is keratin. Feathers belong to natural keratin, a kind of hard protein, which is difficult to be digested and utilized, resulting in a serious waste of protein resources. [0004] Statistics show that in China, the annual output of feathers can be as high as 11 million tons; whil...
