Embodiment 1

[0086] Mice were injected with tumor cells (sc) from tumors expressing high levels of CXCL12 and allowed to develop. After tumors had formed, mice were injected (subcutaneously on the same side as the tumor) with AMD3100 or vehicle once daily for 5 days.

[0087] One to three days after the final dose of AMD3100, mice were injected with a therapeutically effective amount of cisplatin via intraperitoneal injection, and tumor growth assays were performed 18 hours later. Tumor growth in mice was delayed by cisplatin treatment, but recovered quickly after discontinuation of treatment in mice not administered AMD3100. Treatment with AMD3100 prior to treatment with cisplatin is expected to be synergistic such that co-treatment results in a longer delay in tumor growth than cisplatin alone.

Abstract

The invention described herein relates to compositions for treating cancer in a patient, or a tumor cell, by administering an effective amount of AMD3100 over a period of time.

Description

Background of the invention [0001] Cell movement in response to specific stimuli has been observed in prokaryotes and eukaryotes. Cell motility observed in these organisms has been divided into three categories: chemotaxis, or movement of cells along a gradient of increasing chemical concentration; negative chemotaxis, which is defined as movement down a gradient to a chemical stimulus; and chemical Chemokinesis, or increased random movement of cells induced by a chemical agent. [0002] Chemotaxis and chemical activation have been observed to occur in mammalian cells in response to a class of proteins known as chemokines. In addition, chemical repellent or fugetactic activity has been observed in mammalian cells. For example, some tumor cells secrete chemokines in concentrations sufficient to expel immune cells from the tumor site, thereby reducing the ability of the immune system to target and eradicate tumors. Metastatic cancer cells can use similar mechanisms to evade t...
