Problems solved by technology

The secondary stress signals of drought and salt stress are caused by water shortage or salt enrichment, including oxygen stress, destruction of cellular components (such as membrane lipids, proteins, and nucleic acids), and metabolic dysfunction, etc., and the mechanism is more complicated.

Embodiment 1

[0025] 1. Materials

[0026] The methods used in this embodiment are conventional methods known to those skilled in the art unless otherwise specified, and the reagents and other materials used, if not otherwise specified, are commercially available products.

[0027] 2. Method

[0028] 2.1 Cloning of maize ZmDi19-9 gene

[0029] 2.1.1 Design of primers

[0030]According to the published data on the website of the Maize Genome Database, the CDS sequence of the maize ZmDi19-9 (GRMZM2G014066_T01) gene was obtained. The design of upstream and downstream primers requires Primer Premier 5.0 software, combined with the subsequent use of the overexpression vector pCAMBIA1301a vector polycloning site, and through the restriction enzyme site analysis website (http: / / nc2.neb.com / NEBcutter2 / ) to determine the available restriction sites of the gene, and finally determine the introduction of KpnI (GGTACC) in the upstream and BamHI in the downstream (GGATCC) and their restriction site p...

Abstract

The invention discloses a corn stress resistance relevant gene ZmDi19-9 and application thereof. The nucleotide sequence of the gene is disclosed in SEQ ID NO.1 and is a new plant stress resistance relevant gene. The gene has a highest expression level in stems, the contents in roots and leaves take a second place, and the contents in tassels are the lowest. A subcellular positioning experiment result displays that the ZmDi19-9 protein is the protein positioned in a cell nucleus and a cytomembrane. A transcriptional activity experiment result indicates that the ZmDi19-9 protein has transcriptional self-activation activity. The discovery of the gene enriches the resources of the stress resistance relevant gene so as to perform an important meaning for the breeding of the corn stress resistance new variety. Compared with an existing discovered corn stress resistance relevant gene, the corn stress resistance relevant gene disclosed by the invention has better stress resistance ability.

Description

technical field [0001] The present invention relates to the technical field of genetic engineering, in particular to a corn stress resistance-related gene ZmDi19-9 and its application. Background technique [0002] Corn has become one of the most important food crops in the world. As the largest food crop in my country, corn is planted in a vast area and has a rich yield. The annual corn planting area in the world can reach more than 2 billion mu, and the total output can reach 1 billion tons. However, more than 50% of the corn planted in my country is located in the northeast, southwest and northwest regions that lack water, and corn is seriously affected by abiotic adversities such as drought. In addition, soil salinization under abiotic stress will also seriously affect the yield of corn. Some scholars predict that by 2050, more than half of the world's soil will be salinized. It can be seen that it is urgent to cultivate high-yield and stress-resistant maize varieties. ...
