Embodiment 1

[0029] A method for assessing the impact of ionic liquids on glucose metabolism, said method comprising the following steps:

[0030] HepG2 cells in the logarithmic growth phase were digested with trypsin, and the digested HepG2 cells were counted with a cell counting plate and diluted with cell culture medium to a cell concentration of 1×10 5 individual / mL. Take a 96-well plate, in which, add 100 μL of cell solution to each well of 48 wells (each 6 wells are randomly divided into a group of 8 treatment groups), take another 6 wells to add only cell culture medium as a control group, and add phosphoric acid to the remaining wells. Salt buffer solution (PBS) was used to prevent bacterial contamination, and cultured in a cell incubator for 24 hours to allow the cells to adhere to the wall. After the HepG2 cells were cultured and attached to the wall, the original medium was aspirated, and 100 μL of exposure medium with exposure concentrations of 0, 0.5, 1, 2, 4, 8, 16, and 32 m...

Abstract

The invention relates to the field of environmental toxicology models, in particular to an assessment method of influence of ionic liquid on glycometabolism. According to the assessment method disclosed by the invention, HepG2 cells are used as experiment cells, repeated exposure treatment is performed on the HepG2 cells by the ionic liquid namely 1-butyl-3-methylimidazolium chloride, then the treated HepG2 cells are subjected to detection of glycogen content and glycometabolism related gene expression, and an assessment method of the ionic liquid on glycometabolism toxicity, which is simple and convenient to operate and short in period is provided. The assessment method disclosed by the invention has comprehensiveness and universality on detection of genes corresponding to relevant rate-limiting enzymes in each key stage of glycometabolism. The assessment method provided by the invention can comprehensively and accurately assess the glycometabolism toxicity level of the ionic liquid, and a basis is provided for determining the standard of ecological environment pollutants.

Description

technical field [0001] The invention relates to the field of environmental toxicology models, in particular to a method for evaluating the influence of ionic liquids on sugar metabolism. Background technique [0002] Ionic Liquid (Ionic Liquid) refers to a class of liquid composed of organic cations and inorganic or organic anions. Compared with traditional volatile toxic solvents, ionic liquids have good chemical stability, non-volatility, non-flammability, almost It has no advantages such as vapor pressure, and is a "green solvent" widely used in organic synthesis, biocatalysis, electrochemistry and other fields. In recent years, researchers have found that ionic liquids have biological toxicity by using experimental models such as bacteria, fungi, mice, fish, and mammalian cell lines. With the increasing application of ionic liquids, their environmental risks have become a research hotspot. [0003] Human hepatoma cell line (HepG2) is a type of cell line isolated from h...
