Abstract

The invention provides an ENO1 gene promoter region methylation site and application thereof in preparation of a kit for early diagnosis and prognosis evaluation of esophageal cancer, and belongs to the technical field of disease diagnosis and prognosis rating. A methylation site of an ENO1 gene promoter region is as shown in SEQ ID No.1, DNA of esophageal squamous carcinoma and para-carcinoma normal sample tissues is extracted, the methylation frequency of a CpG site of the ENO1 gene promoter region is detected by adopting flight mass spectrometry after the DNA is modified by hydrosulfite, and meanwhile, expression of ENO1 protein in the esophageal squamous carcinoma and para-carcinoma normal tissue samples is detected by adopting immunohistochemistry; research finds that an ENO1 gene promoter region in esophageal squamous carcinoma is low in methylation and ENO1 protein is high in expression, patients with low methylation are poor in prognosis, and ENO1 can promote glycolysis level of esophageal carcinoma cells, promote proliferation of the esophageal carcinoma cells and inhibit apoptosis of the esophageal carcinoma cells; the invention relates to a reagent for early diagnosis and prognosis evaluation of esophageal squamous carcinoma by detecting the methylation level of an ENO1 gene.
