Embodiment 1

[0293] Example 1. Identification of LD block C region related to prostate cancer

[0294] Patients included in genetic studies

[0295] The population based on the list of all prostate cancer patients diagnosed with prostate cancer in Iceland from 1955 to 2005 forms the basis of the present invention. Patients who were onset from 2001 were invited to join the study. As of October 2006, blood samples had been collected from 1,564 prostate cancer patients. 1455 prostate cancer patients and 4182 control individuals in this population were genotyped using Illumina 317K Bead chips.

[0296] Statistical methods for association and haplotype analysis

[0297] For the association between a single marker and the disease, Fisher's exact test was used to calculate the two-sided P-value for each single allele. When displaying the results, we used allele frequencies instead of SNPs and haplotype carrier frequencies. Haplotype analysis is performed using a computer program called NEMO (NEsted M...

Embodiment 2

[0363] Example 2: Confirmation of association with prostate cancer in several groups

[0364] Other analyses further support the presence of variants associated with prostate cancer on chromosome 8q24. As shown in Table 7, both rs16901979 allele 1 and HapC were found to be associated with prostate cancer in two groups of Caucasian descent and one group of African descent.

[0365] Repeated research on the association between single markers and haplotypes in LD block C and prostate cancer. The group contained samples of Caucasians from Chicago (Northwestern University) and Spain (Zaragoz University Hospital), and a group of African Americans from Michigan (University of Michigan). The allele encoding of SNPs is as follows: 1=A, 2=C, 3=G, 4=T, X=any allele.

[0366] Table 7. Correlation between SNP markers on Chr8q24 and prostate cancer (PrCa) in Spain and the United States

[0367] Shows the alleles of markers rs16901979 and SNPs in the same LD block of 8q24.21, the corresponding nu...

Abstract

A region on chromosome 8q24.21 has been demonstrated to play a major role in particular forms of cancer. It has been discovered that certain markers and haplotypes are indicative of a susceptibility to particular cancers, including prostate cancer. Diagnostic applications for identifying a susceptibilty to cancer using these markers and haplotypes are described.

Description

Background of the invention [0001] Cancer, the uncontrolled growth of malignant cells, is a major health problem in the modern medical era, and in developed countries, it is one of the leading causes of death. In the United States, one in four deaths is caused by cancer (Jemal, A. et al., CA Cancer J. Clin. 52:23-47 (2002)). [0002] In the past few decades, the incidence of prostate cancer has risen sharply. Currently, prostate cancer has become the leading cause of death in the United States and Western Europe (Peschel, RE and JW Colberg, Lancet 4:233-41 (2003); Nelson, WG et al., N. Engl. J. Med. 349(4): 366-81 (2003)). In industrialized countries, prostate cancer is the most frequently diagnosed non-cutaneous malignant tumor among men. In the United States, 1 in 8 men will develop prostate cancer during their lifetime (Simard, J., etc., Endocrinology 143 ( 6): 2029-40 (2002)). Although environmental factors such as dietary factors and lifestyle-related factors contribute to...
