Problems solved by technology

So far, these methods are very limited in terms of the effect and performance improvement of gelatin modification, because the number of main side chain amino or carboxyl groups available for chemical modification of gelatin itself is quite limited

Embodiment 1

[0030] Tetrahydrofuran solvent 1L, gelatin particles (300g), gelatin grinding to make the particle size more refined to 80 mesh, to meet the solid-liquid phase reaction conditions. Dissolve 3% (30g) succinic anhydride in tetrahydrofuran solvent, suspend and swell the ground gelatin particles in the mixed liquid phase, add 0.01% (0.1g) nicotinamide as catalyst, 0.01% (0.1g) poly Ethylene glycol is used as a cocatalyst, and the reaction temperature is controlled at 50°C. The reaction is carried out in a colloid mill, and the reaction is ground for 4 hours to obtain modified gelatin. After the reaction, most of the tetrahydrofuran solvent was removed by filtration. For a small amount of residual solvent, due to the low boiling point of tetrahydrofuran, the residual solvent was completely removed by vacuuming. The degree of modification of the gelatin is measured by a formaldehyde titration method, and the modified gelatin with a modification degree greater than 90% and no solvent...

Abstract

The invention relates to a preparation method of a modified gelatin, which comprises the following steps of: (a) grinding large-particle gelatin to ensure that the gelatin is thinned to have a particle size of over 80 meshes; and (b) dissolving an acylation agent into tetrahydrofuran for mixing, putting the gelatin particles, which are thinned by grinding, into a blended liquid phase through suspension swelling, and performing acylation reaction by taking nicotinic acid, nicotinamide or acidylated histidine as a reaction catalyst and polyglycol as a cocatalyst to obtain the modified gelatin. Compared with the former unmodified gelatin, part of amino groups of the modified gelatin are closed, and the modified gelatin is grafted with acrylate of which one end still has free carboxyl groups, so multi-aspect properties of the gelatin are changed and improved, and after modification, the water solubility and swelling property of the gelatin are obviously increased, and the stability is also increased to a certain extent. Besides, a solvent, a modifier and a catalyst in the process of the reaction are nontoxic, so the serviceable range of the modified gelatin is enlarged, the reaction condition is mild and safe, and the modified gelatin is suitable for industrial production and has a good application prospect.

Description

technical field [0001] The invention relates to a preparation method of modified gelatin. Background technique [0002] Gelatin is the denatured product of collagen under the action of acid, alkali, enzyme or high temperature, and it is a polymer of natural polypeptide. Gelatin is derived from collagen, which exists in the connective tissue (including soft tissue, animal skin, bone and tendon) of animals (pigs, cattle, etc.), and gelatin protein contains 18 kinds of amino acids. Therefore, gelatin has excellent biocompatibility and high nutritional value. [0003] Gelatin has the characteristics of gelation, and the greater the molecular weight of gelatin and the longer the molecular chain, the more conducive to the formation of a network structure; gelatin has good stability, relatively stable at room temperature and in a dry state, and can be placed for several years . Because gelatin has many of the above advantages, the development of gelatin-based biomaterials has be...
