Embodiment 1

[0031] Embodiment 1, a kind of induction method that can promote the salvianolic acid (mainly comprising rosmarinic acid and salvianolic acid B) accumulation in the hairy root of Salvia miltiorrhiza miltiorrhiza, carries out the following steps successively:

[0032] 1), initial cultivation of salvia miltiorrhiza hairy roots:

[0033] In an ultra-clean bench, weigh 0.2 g of Salvia miltiorrhiza root, put it into 50 ml of 6,7-V medium (100 ml Erlenmeyer flask, pH: 5.8), shaker at 110 rpm, and culture in the dark at 25°C for 18 sky.

[0034] 2), the continued cultivation of salvia miltiorrhiza root:

[0035] After the Salvia miltiorrhiza root was cultured for 18 days, sodium nitroprusside was added to make the final concentration of sodium nitroprusside in the 6,7-V medium 100 μM; culture was induced for 6 days at 110 rpm, 25° C., and in the dark.

[0036] After 6 days of induction, the hairy roots of Salvia miltiorrhiza were collected, rinsed with distilled water, blotted dry ...

Abstract

The invention discloses an inducing method capable of promoting danshinolic acid accumulation in a hairy salvia miltiorrhiza root. The method comprises the following steps: (1) carrying out primary culture on the hairy salvia miltiorrhiza root, namely inoculating 0.15-0.2g of hairy salvia miltiorrhiza root in each 50mL of a 6,7-V culture medium, carrying out dark culture under the culture conditions of 100-150rpm and 24-26 DEG C for 17-19 days; and (2) further culturing the hairy salvia miltiorrhiza root, namely adding sodium nitroprusside, controlling the final concentration of the sodium nitroprusside in the 6,7-V culture medium to be 90-110muM, and carrying out the dark culture under the culture conditions of 100-150rpm and 24-26 DEG C for 5-7 days. According to the method disclosed by the invention, the accumulation of the danshinolic acid (mainly including rosmarinic acid and danshinolic acid B) in the hairy salvia miltiorrhiza root can be promoted.

Description

technical field [0001] The invention relates to an induction method capable of promoting the accumulation of rosmarinic acid and salvianolic acid B in hairy roots of Salvia miltiorrhiza. Background technique [0002] Salvia miltiorrhiza (Salvia miltiorrhiza) is the root and rhizome of Salvia miltiorrhiza, which has the effects of promoting blood circulation and removing blood stasis, clearing heat and calming nerves, nourishing blood and nourishing qi. Salvianolic acid is one of the active ingredients of Danshen, mainly including rosmarinic acid and salvianolic acid B. Due to its outstanding performance and non-toxic side effects in the treatment of cardiovascular diseases, the application and research of Danshen are becoming more and more extensive. [0003] Currently, the hairy root of Salvia miltiorrhiza can be cultured in 6,7-V medium. Generally, 0.15-0.2 g of fresh hairy roots are weighed and inoculated into 50 mL of 6,7-V medium for dark culture. The hairy root of S...
