The blood ammonia determination reagent comprises a buffer solution, a preservative and a stabilizing agent, and further comprises leucine dehydrogenase, alpha-ketonic acid NADH (Reduced Form of Nicotinamide-adenine Dinucleotid), beta-hydroxybutyric acid and beta-hydroxybutyrate dehydrogenase, wherein the enzyme activity unit ratio of beta-hydroxybutyrate dehydrogenase to leucine dehydrogenas is 1:10-2,000. According to the blood ammonia determination reagent provided by the invention, reduced coenzyme I(NADH) is oxidized into oxidized coenzyme I (NAD+) by applying a leucine dehydrogenase reaction system; the content of blood ammonia is directly reflected by determining the decrease rate of the reduced coenzyme I in the dominant wave of 340 nm. The sensitivity of the blood ammonia determination reagent can be ensured, and the stability of the NADH can also be ensured by adjusting the proportion of the beta-hydroxybutyrate dehydrogenase to the leucine dehydrogenase to be 1:10 to 2,000 through an experiment.
