A method for determining the acute and chronic toxicity of drugs to hornet mites

A technology of vampire mite and medicine, which is applied in animal husbandry and other fields, can solve the problems of inability to measure the chronic toxicity of vampire mite, high mortality rate of vampire mite, and unreliable results, so as to shorten the research, development and utilization cycle and control the test conditions , the result is accurate and credible

Problems solved by technology

The disadvantage of the indoor toxicity test method is that due to factors such as lack of food for the bee mite, the mortality rate of the bee mite in the control group is high, the result is not credible, and the chronic toxicity of the drug to the large bee mite cannot be determined.

Embodiment 1

[0044] Embodiment 1, the assay method of the toxicity of medicine to the giant bee mite

[0045] Preparation before the test: Western honey bee (Apis melifera) bee colony (healthy colony, infected bee mite colony), incubator, ultra-clean bench, balance, magnetic stirrer, mixing oscillator, thermo-hygrometer, pipette, ultra-clean Table heater, cold light source lighting, egg laying controller, insect transfer needle, tweezers, 48-hole larvae culture plate, larvae culture cup, bee mite collector (petri dish with cover), small brush for mites transfer, paraffin film, Honeycomb insulation box, latex gloves, masks, royal jelly, glucose, fructose, yeast extract, potassium sulfate, sodium chloride, double distilled water, drugs to be tested. Unless otherwise specified, the drugs and equipment used in this example can be obtained from commercial sources.

Abstract

The invention discloses a method for measuring acute and chronic toxicity of a drug against varroa jacobsoni. The method comprises the following steps: gently spraying the to-be-measured liquid onto the surfaces of healthy bee larvae; transferring the bee larvae from the inside of a larva culture plate into a bee pupa culture cup with the liquid-infiltrated filter paper piece at the bottom; collecting the to-be-measured varroa jacobsoni; introducing a varroa jacobsoni into the bee pupa culture cups of each treatment group and control group, and sealing the cup rims of the bee pupa culture cups with ventilated transparent films to prevent the introduced varroa jacobsoni from escaping the bee pupa culture cups; putting the bee pupa culture cups into an incubator for culturing, wherein the temperature is 34.5 DEG C, and the relative humidity is 75%; counting the deaths of varroa jacobsoni 48h or 72h later, calculating the death rate and analyzing the acute toxicity of the drug against varroa jacobsoni; or continuously recording the survival conditions of the varroa jacobsoni every day, counting the survival time of each varroa jacobsoni, and analyzing the chronic toxicity of the drug against varroa jacobsoni. The method disclosed by the invention has the advantages of controllable experiment conditions, high survival rate of the varroa jacobsoni, long survival time and accurate and credible result.

Description

technical field [0001] The invention relates to a method for measuring the toxicity of medicines to insects, in particular to a method for measuring the acute and chronic toxicity of medicines to venomous bee mites. Background technique [0002] Honey bees are important economic insects with great economic and ecological value. Honey bees can not only provide beneficial bee products but also pollinate plants. The bee mite is the most serious bee enemy that harms bee industry. [0003] The large bee mite is the most serious bee enemy that endangers the beekeeping industry in the world. Due to the long-term use of pyrethroid acaricides, the large bee mite has developed serious drug resistance, and the screening of new high-efficiency green acaricides is imminent. However, there is a lack of standard methods for the determination of the toxicity of acaricides to the venomous mite in the world. At present, two methods are generally used, one is colony virulence assay, and the o...
