Abstract
The invention discloses a method for preserving arbuscular mycorrhizal fungi. The moisture in propagules of the arbuscular mycorrhizal fungi is fully reduced before cryopreservation of strains. The method comprises the following steps: ventilating and drying at normal temperature, and reducing the majority of moisture in a matrix material so that the moisture content is reduced to below 8%; and then reducing the moisture content in the propagules by utilizing silica gel drying in a glass dryer so that the moisture content of a matrix is reduced to below 5%. The moisture content of the propagule material is reduced by two steps so that the moisture content in arbuscular mycorrhizal fungi spores and mycelial cells is slowly reduced, thus being beneficial to maintain the vitality of the strains and simultaneously avoiding the damage of ice crystals to the cells during the rapid freezing. Through the experimental comparison, the activity of strains of the arbuscular mycorrhizal fungi can be effectively preserved. Compared with a pot culture inoculating method and an cryopreservation method, the method does not occupy a culture greenhouse, does not need the regular artificial inoculation and does not need a strain protection agent and a liquid nitrogen preservation material, the operation is simple, and the material resources and manpower are saved.
