The method comprises the following steps: 1) determining the position of a central axis of a cylinder, establishing a coordinate system with the central axis of the cylinder being an axis, and calibrating coordinates of cylinder center datum mark points on the cylinder through a datum transformation method; 2) setting cylinder roundness measurement control points and carrying out measurement; and 3) establishing a cylinder roundness measurement local area network and measuring the roundness of the cylinder. An absolute laser tracker is introduced, so that under the condition of narrow and small space and hard datum point measurement, the roundness of super-large cylinders, such as a hydro-generator rotor and a stator, can be measured, and measurement accuracy and reliability are improved.
