The invention discloses a preparation method of a vaccinium myrtillus extract, which comprises the following steps of adding an acidized ethanol solution into fruits of vaccinium myrtillus, stirring and extracting, after the extracting is finished, stirring and adding a saturated sodium citrate solution, centrifuging, discarding filter residues, adding water into a concentrated solution and loading the concentrated solution on a macroporous adsorbent resin column, washing with a citric acid solution, desorbing by an ethanol solution, concentrating feed liquid until non-alcohol, and drying to obtain the vaccinium myrtillus extract. The preparation method is simple and convenient in process, easy to operate and convenient for industrialized production; the anthocyanin (accounting by cyanidin 3-0-glucoside chloride) content in the prepared vaccinium myrtillus extract is greater than or equal to 36%, and the total anthocyanin (accounting by cyanidin chloride) content is less than or equal to 1.0%, which accord with the requirements of United States pharmacopeia and European pharmacopoeia; the content of radionuclide <137>Cs in the vaccinium myrtillus extract is low, and is less than 10Bq/kg, which accord with the national standard of food safety (GB 14883.10-2016).
