Problems solved by technology

The Institute of Deep Sea Science and Engineering, Chinese Academy of Sciences used a self-made trap to trap a small amphipod with a body length of less than 1cm at a depth of 10908m in the Mariana Trench. At present, there is no report on the mitochondrial genome of this species.

Embodiment 2

[0086] 1. Sample collection: Use a trap to collect abyssal amphipods at 10908m in the Mariana Trench, and store the collected samples in a -80°C refrigerator.

[0087] 2. DNA extraction: use the SQ Tissue DNA Kit of OMEGA biotek to extract the genome of the small amphipods of the abyss, the specific method is as follows:

[0088] 1) Take 3 mg of small amphipod tissue frozen samples, put them into a 1.5 ml sterilized centrifuge tube with 300 μl of WTL buffer, and cut the tissue into pieces with scissors after cleaning and burning;

[0089] 2) Add 5μl 20mg / ml proteinase K to the centrifuge tube, vortex for 15s; place at 60°C for more than 6 hours, and briefly centrifuge to remove the water droplets hanging on the tube wall;

[0090] 3) Cool the centrifuge tube to room temperature, add 100 μl of PCP buffer, vortex for 30 seconds, and ice-bath for 5 minutes;

[0091] 4) Centrifuge at a speed of 13000g for 3 minutes, transfer the supernatant to another sterile 1.5ml centrifuge tub...

Abstract

The invention discloses a mitochondrial genome sequence derived from abyssal new species. The genome sequence is shown in SEQ ID NO:1. The invention also provides a method for acquiring the genome sequence. The mitochondrial genome sequence of small abyssal amphipoda, which is obtained with the method, is a precious deep-sea organism gene resource, and composition and structure analysis of the mitochondrial genome sequence can lay important molecular biology theoretical foundation for system development of amphipoda, expansion of a germplasm resource bank and an adaption mechanism to the abyssal environment.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to a mitochondrial genome sequence derived from an abyssal new species. Background technique [0002] Mitochondria are important organelles for biological oxidation and energy supply in cells. The mitochondrial genomic DNA of eukaryotes is usually a naked covalently double-stranded closed circular molecule consisting of two chains, a heavy chain and a light chain. Mitochondrial genomic DNA generally consists of 37 coding genes and non-coding control regions, of which coding genes include: 13 hydrophobin genes, 22 tRNA genes and two rRNA genes. Compared with nuclear genomic DNA, mitochondrial genome has the characteristics of small molecular weight, maternal inheritance, high copy number, low recombination rate, and fast evolution rate. It has been widely used in phylogeny and species identification, genetics, biogeography, and conservation biology. At the same time, for endangered spe...
