Abstract

The invention discloses a method for preparing two main anthocyanin standard substances in cloudy juice of vitis davidii by utilizing high-speed counter-current chromatography. The method comprises the following steps: enriching the cloudy juice of vitis davidii by adopting a macroporous resin, eluting with ethanol, performing freeze drying to obtain a crude sample, performing high-speed counter-current separation on the crude sample with water in a solvent system, and separating to prepare anthocyanin standard substances, wherein the anthocyanin standard substances are respectively malvidin-[3-O-glucose-]-[5-O-glucose] with purity of 95.8% and malvidin-[3-O-beta-glucosyl-6-coumaroyl]-[5-O-beta-D-glucose] with purity of 92.2%. The yield of the two anthocyanin standards prepared from the cloudy juice of vitis davidii is respectively 122.77mg / L and 78.09 mg / L. A method for separating anthocyanin and polyphenol in the cloudy juice of vitis davidii by high-speed counter-current chromatography is established, and the method for preparing high-purity standard substances can meet large-scale preparation of natural anthocyanin of the vitis davidii.
