Embodiment 1

[0024] Such as Figure 1-5 As shown, this embodiment provides a model construction method and effect verification that can predict the survival possibility of bladder cancer patients.

[0025] 1.1 Data download and preprocessing

[0026]Download Fragments Per Kilobase of transcript perMillion mapped reads (FPKM) standardized RNA sequencing dataset and clinical data from The Cancer Genome Atlas (TCGA), which contains 411 tumor samples and 19 non-tumor samples. After normalization, the R package "org.Hs.eg.db" was used to convert the probes into gene names, and for genes corresponding to multiple probes, the probe with the largest average expression value was selected.

[0027] 1.2 Screening of prognostic-related DNA repair genes

[0028] To construct a DNA repair-related prognostic model, 563 DNA repair-related genes (DRRG) were obtained from Gene Set Enrichment Analysis (GSEA) (https: / / www.gsea-msigdb.org / gsea / index.jsp). The R package "limma" was used to analyze DRRG diffe...

Abstract

The invention belongs to the technical field of bioengineering and tumor markers, and relates to a prognostic model for the overall survival rate of bladder cancer patients, in particular to a prognostic model that can predict the overall survival rate of bladder cancer patients based on 12 DNA repair-related genes. The present invention establishes a prognosis model with 12 DRRGs, and divides bladder cancer patients into high-risk and low-risk groups. Risk scores for bladder cancer patients in the training cohort were significantly associated with OS (P < 0.001; HR = 6.3 [4.1, 9.8]). ROC curve analysis showed that in 1-year, 3-year and 5-year follow-up, the AUCs were 0.763, 0.735 and 0.735, respectively. The predictive performance has been verified on the test set.

Description

technical field [0001] The invention belongs to the technical field of bioengineering and tumor markers, and relates to a prognostic model for the overall survival rate of bladder cancer patients, in particular to a prognostic model that can predict the overall survival rate of bladder cancer patients based on 12 DNA repair-related genes. Background technique [0002] Bladder cancer (BLCA) is the most common malignant tumor of the urinary system, and its morbidity and mortality are high. About 75% of bladder cancer patients are non-muscle invasive bladder cancer (NMIBC) at first diagnosis, and about 25% of patients are Muscle invasive bladder cancer (MIBC) or metastatic disease. NMIBC is usually treated locally with intravesical chemotherapy or immunotherapy combined with TURBT, however most NMIBC relapse within 6–12 months, and 10%–15% of patients may progress to invasive or metastatic disease. Overall, the 5-year survival rate for all stages of bladder cancer still does n...
