Embodiment 1

[0036] 1. Serum Sample Collection

[0037] Before the collection of serum samples, all volunteers included in the study signed the informed consent.

[0038] Serum samples were collected under the same conditions: 74 cases of diabetic patients without retinopathy as the control group and 234 cases of diabetic patients with retinopathy (including 66 cases of stage 1 mild diabetic retinopathy group and stage 2 moderate diabetic retinopathy group). 60 cases, 57 cases of stage 3 severe diabetic retinopathy group and 51 cases of stage 4 proliferative diabetic retinopathy group) were used as the disease group. After collection, the samples were stored in a -80°C refrigerator for future inspection.

[0039] 2. Analysis method

[0040] 2.1 Serum sample pretreatment

[0041]Thaw the sample at 4°C, take 50 μL of serum, add 200 μL of methanol extract containing internal standard compounds D4-succinic acid, D5-phenylalanine and tridecanoic acid (concentrations are 5 μg / mL), vortex for 3...

Embodiment 2

[0053] 1. Serum Sample Collection

[0054] Before the collection of serum samples, all volunteers included in the study signed the informed consent.

[0055] Serum samples were collected under the same conditions: 37 cases of diabetic patients without retinopathy as the control group and 116 cases of diabetic patients with retinopathy (including 33 cases of stage 1 mild diabetic retinopathy group and 2 cases of moderate diabetic retinopathy group). 30 cases, 28 cases of stage 3 severe diabetic retinopathy group and 25 cases of stage 4 proliferative diabetic retinopathy group) were used as the disease group. After collection, the samples were stored in a -80°C refrigerator for future inspection.

[0056] 2. Analysis method

[0057] With embodiment 1.

[0058] 3. Serum test results and auxiliary predictive analysis

[0059] The results of the verification set in Example 2 are basically consistent with the results of the discovery set in Example 1. In the diabetic retinopathy...

Abstract

The invention relates to a novel application of small molecule metabolites ethanolamine, fumaric acid, hypotaurine and inositol in a serum sample as a combined marker in preparation of a kit for diagnosing whether a diabetic has retinopathy or not. The invention also relates to a kit for detecting retinopathy in diabetic patients, which is used for calculating the variable Prob value of the combined marker and judging the cut-off value (cut-off value) on the basis of a binary logistic regression equation by detecting the respective relative concentrations of the combined marker in the serum of the diabetic patients, and judging whether the diabetic suffers from retinal complications or not. The kit can realize high-sensitivity and high-efficiency detection of several small molecule metabolites involved in the invention, and has the characteristics of low detection cost and good repeatability. The method can be applied to auxiliary diagnosis of retinal complications in diabetic people, has a good effect in early diagnosis of diabetic retinopathy, and has a good application prospect.

Description

technical field [0001] The present invention relates to the application of a new small molecule combined marker in the diagnosis of diabetic retinopathy. It belongs to the fields of analytical chemistry, biochemistry and clinical medicine. Background technique [0002] Diabetes is one of the most common global metabolic diseases and its incidence has been increasing year by year. Diabetic retinopathy is one of the most common microvascular complications in diabetic patients, and it is also the main cause of visual impairment and blindness in adults [Reference 1: Nentwich M M, Ulbig M W. Diabetic retinopathy-ocular complications of diabetes mellitus[J]. World journal of diabetes, 2015, 6(3): 489.]. The presence and grade of diabetic retinopathy can be assessed clinically by performing a fundus examination after pupil dilation (mydriasis) to visualize the entire retina. Diabetic retinopathy can be divided into background retinal mutation stage and proliferative retinopathy ...
