The invention discloses a
cysteine sulfinate decarboxylase
mutant with improved enzymatic activity, and belongs to the field of
gene engineering and
enzyme engineering. According to the present invention,
cysteine sulfinate decarboxylase derived from
aedes aegypti is adopted as a parent
enzyme, and site-
directed mutagenesis modification is performed on the
amino acid sequence of the
cysteine sulfinate decarboxylase through
rational design so as to obtain the
mutant capable of significantly improving the
enzyme activity; according to the
amino acid sequence of the
mutant, compared with a parent enzyme,
threonine at the 410th site is replaced with
proline, and
threonine at the 498th site is replaced with
tryptophan. A determination result shows that the
specific enzyme activity of the two-site mutant is 4.19 times that of a parent enzyme. The cysteine
sulfinic acid decarboxylase mutant can efficiently catalyze cysteine
sulfinic acid to generate
hypotaurine through
decarboxylation, provides a key enzyme preparation for green and efficient synthesis of
taurine, and has important application prospects in the fields of
food additive preparation, medical intermediate synthesis, biological catalytic
reaction system construction and the like. And a foundation is laid for industrial application of the cysteine sulfinate decarboxylase.
