[0191] Anti-NGF monoclonal antibody combined with NSAID in the treatment of postoperative pain

[0192] We evaluated the efficacy of an anti-NGF antibody in combination with an NSAID (ibuprofen) using a pain model simulating postoperative pain. For each experiment, 16 male adult Sprague Dawley rats (Harlan; Indianapolis, IN) weighing 200 to 200 g were fed ad libitum with food and water in a room under normal light conditions for at least 1 week prior to use. After 2 hours of acclimatization in the laboratory prior to surgery, rats were divided into two groups: one group received antibody 15 hours before surgery and the other received vehicle (5% dextrose / 0.45% saline USP) at that time. Anti-NGF antibody antagonist 911 was administered at 1 mg / kg body weight (see, Hongo et al., Hybridoma 19:215-227 (2000)). All animals were administered ibuprofen at various concentrations (sc) of 10, 200 and 300 mg / kg 24 hours after surgery.

Abstract

The present invention features methods for treating or preventing pain comprising administering an amount of a nerve growth factor antagonist (such as an anti-NGF antibody) and an amount of an NSAID such that together they provide effective pain relief. The invention also features compositions comprising a nerve growth factor antagonist and an NSAID and kits containing the same.

[0001] Cross References to Related Applications [0002] This application claims the benefit of U.S. Provisional Application Serial No. 60 / 448,823, filed February 19, 2003, and U.S. Provisional Application Serial No. 60 / 448,853, filed February 19, 2003, the contents of which are incorporated by reference in their entirety . field of invention [0003] The present application relates to methods and compositions for the prevention and treatment of pain by administering to a patient a nerve growth factor antagonist in combination with an NSAID. Background of the invention [0004] Some treatments including the administration of non-steroidal anti-inflammatory drugs (NSAIDs) are currently recommended for pain relief. Administration of NSAIDs has been shown to exhibit pain-reducing features. However, treatment with NSAIDs is known to have disadvantages, including unwanted side effects such as gastrointestinal irritation and renal and hepatotoxicity. Furthermore, NSAIDs, even a...
