[0191] Anti-NGF monoclonal antibody combined with NSAID in the treatment of postoperative pain

[0192] We evaluated the efficacy of an anti-NGF antibody in combination with an NSAID (ibuprofen) using a pain model simulating postoperative pain. For each experiment, 16 male adult Sprague Dawley rats (Harlan; Indianapolis, IN) weighing 200 to 200 g were fed ad libitum with food and water in a room under normal light conditions for at least 1 week prior to use. After 2 hours of acclimatization in the laboratory prior to surgery, rats were divided into two groups: one group received antibody 15 hours before surgery and the other received vehicle (5% dextrose / 0.45% saline USP) at that time. Anti-NGF antibody antagonist 911 was administered at 1 mg / kg body weight (see, Hongo et al., Hybridoma 19:215-227 (2000)).
