Problems solved by technology

As a post-exposure immunization vaccine, antibodies should be produced quickly to prevent the virus from invading the nervous system, but the current domestic and foreign vaccines take a long time (10-14 days) to induce the body to produce neutralizing antibodies with effective protective titers, especially for Patients with a short incubation period or a lesion site close to the central ne...

Embodiment 1

[0032] Preparation of anti-rabies virus recombinant immunotoxin

[0033] 1. Screening of hybridoma cell lines that secrete neutralizing monoclonal antibodies against rabies virus

[0034] Balb / c mice were immunized with the purified international standard strain of rabies virus (CVS strain) for a total of three immunizations with an interval of 15 days. After three immunizations, the serum antibody titer was detected (ELISA titer> 2 7 , Agar diffusion test potency>2 3 ), aseptically isolated mouse spleen cells, fused with mouse myeloma cells, screened positive fusion cells by HAT method, and screened hybridoma cell lines secreting high-affinity neutralizing monoclonal antibodies by ELISA method.

[0035] 2. Construction of Rabies Virus Neutralizing Single Chain Antibody (ScFv) Gene

[0036] Use trizol to extract RNA from hybridoma cell lines that secrete neutralizing monoclonal antibodies, and synthesize light and heavy chain cDNAs complementary to each other. PCR method is used t...

Abstract

The present invention discloses preparation process of rabies virus resisting recombinant immunotoxin. The preparation process includes screening out hybridoma strain capable of generating specific rabies virus resisting monoclonal antibody, extracting hybridoma genome, amplifying antibody variable heavy chain region gene and variable light chain region gene in a PCR process, constituting rabies virus resisting neutral single chain antibody, cleaving the single chain antibody and inserting into expression vector containing Bacillus pyocyaneus exotoxin, transforming colibacillus for prokaryotic express, and purifying the expressed fusion protein. The present invention can neutralize free virus and kill virus infected cell by means of PE cytotoxicity, and may be used in the emergency prevention and treatment of rabies. The present invention is superior to chemically prepared immunotoxin, which has the demerits of low yield, large molecule, great immunogenicity, etc.

Description

Technical field: [0001] The invention relates to an anti-rabies virus immunotoxin, and in particular provides an anti-rabies virus recombinant immunotoxin and a preparation process for emergency prevention and post-exposure treatment of rabies, and belongs to the technical field of the preparation of drugs for prevention and treatment of major zoonoses. Background technique: [0002] Rabies has been recorded as early as the fourth century BC. It is a direct contact and viral infectious disease caused by rabies virus. All warm-blooded animals are susceptible. It is currently an important disease endangering human health. According to statistics from the World Health Organization, 40,000-70,000 people die from rabies every year in the world. The total number of deaths from rabies in my country from 1950 to 2005 was about 102,280. Since January 2006, the number of deaths due to rabies in my country has been on the rise every month. Since May, it has surpassed the long-term leading t...
