Embodiment Construction

[0057] The embodiments of the invention described herein relate to monoclonal antibodies that bind to Ang-2. In some embodiments, the antibody binds Ang-2 and inhibits the binding of Ang-2 to its receptor Tie2. Other embodiments of the present invention include fully human anti-Ang-2 antibodies, and antibody preparations with therapeutic utility. Such anti-Ang-2 antibody preparations preferably have the desired therapeutic properties, including strong binding affinity to Ang-2, the ability to neutralize Ang-2 in vitro, and the ability to inhibit Ang-2 induced angiogenesis in vivo ability.

[0058] One embodiment of the invention includes antibodies that bind and neutralize Ang-2 but not Ang-1. In one embodiment, the antibody binds to both Ang-2 and Ang-1, but only neutralizes Ang-2. In another embodiment, the antibody binds to both Ang-2 and Ang-1, and neutralizes the binding of Ang-1 and Ang-2 to Tie2.

[0059] Embodiments of the invention also include isolated binding fragments ...

Abstract

Antibodies directed to the antigen Ang-2 and uses of such antibodies are described. In particular, fully human monoclonal antibodies directed to the antigen Ang-2. Nucleotide sequences encoding, and amino acid sequences comprising, heavy and light chain immunoglobin molecules, particularly sequences corresponding to contiguous heavy and light chain sequences spanning the framework regions and/or complementary determining regions (CDR's), specifically from FR1 through FR4 or CDR1 through CDR3. Hybridomas or other cell lines expressing such immunoglobin molecules and monoclonal antibodies.

Description

[0001] This application claims priority to the U.S. Provisional Application Serial No. 60 / 638,354 filed on December 21, 2004 and the U.S. Provisional Application Serial No. 60 / 711,289 filed on August 25, 2005, which are incorporated herein by reference. Invention field [0002] The present invention relates to monoclonal antibodies against Angiopoietin-2 (Ang-2) and applications of these antibodies. More particularly, the present invention relates to fully human monoclonal antibodies against Ang-2 and the applications of these antibodies. The invention also relates to hybridomas or other cell lines expressing these antibodies. The antibodies can be used to diagnose and treat diseases related to the activity and / or overproduction of Ang-2. Background of the invention [0003] Angiogenesis is the process of forming new capillaries from existing blood vessels. It is a basic component of embryogenesis, normal physiological growth, repair and tumor expansion. Although there are many fa...
