Embodiment 1

[0014] 1. Materials and methods:

[0015] 1. Pure seeding and germination:

[0016] On October 13, 2008, the harvested Acer yangbiense seeds were first dried in the shade, and after rubbing to remove the seed wings and sundries, the pure seeds were put into 0.05% potassium permanganate solution for 10 minutes, and then sterilized with 80 % Zinc (Zineb) WP seed dressing. Germination adopts 4 treatments: 1) wet sand seed dressing and place in 3-8°C environment; 2) wet perlite seed dressing and place in 5°C environment; 3) wet perlite seed dressing and place in 0°C environment; 4) wet pearl After the rock seed dressing, put it in a -18°C environment. Take out the seeds twice a month, check the water condition, add appropriate amount of water and mix well according to the situation, and then place it in an environment of 15-20°C for 8 hours for variable temperature treatment.

[0017] 2. Sowing:

[0018] On February 10, 2009, it was found that the above-mentioned 1) method had...

Embodiment 2

[0031] 1. Pure seeding and germination:

[0032] The harvested Acer yangbiense (Acer yangbiense) seeds were first dried in the shade, rubbed to remove seed wings and sundries, and the pure seeds were put into 0.05% potassium permanganate solution for 10 minutes for disinfection, and then 80% Zineb (Zineb ) WP seed dressing. For germination, use wet perlite to dress the seeds and place them in an environment of 5°C. During the germination process, take out the seeds twice a month, check the water condition, add appropriate amount of water and mix well according to the situation, and place them in an environment of 15-20°C8 hours for variable temperature treatment.

[0033] 2. Sowing:

[0034] After the seeds germinate, sow them in time. The sowing substrate is - raw red soil (below the surface soil): rotten leaf soil (decomposed, sieved) = 4:1; Ph value is 6.8, which is weakly acidic. The seeds are sown in plastic baskets of 34×25×8cm. The specific method is: put a filter l...

Abstract

The acer yangbiense is regarded as one of the rarest or endangered species in the world, so a nursing method thereof and a population quantity enlarging method thereof are the basic methods for saving the species. The selected plants of the acer yangbiense which are cultured by the invention are researched in the following aspects of conservation biology, environmental biology, and the like, to provide material foundation, scientific basis and technical support for leading the acer yangbiense back to nature and realizing species reconstruction. The invention comprises the steps of breaking the dormancy of the acer yangbiense seedlings; firstly, sterilizing with 0.05% potassium permanganate solution for 10 minutes; secondly, putting the seedlings under the environment of 5 DEG.C after seeddressing with 80% Zineb wettable powder; putting the seedlings under the environment of 15 DEG.C-20 DEG.C for eight hours twice a moth; germinating; processing for 4 months; and seeding into a growing media with turf: perlite: peat: red earth: organic fertilizer=1:1:1:1:1 after the seedlings germinate.

Description

Technical field: [0001] The invention relates to the field of biotechnology, in particular to a method for raising seedlings and cultivating seeds of the rare and endangered plant Acer japonica. Background technique: [0002] Acer yangbiense (Acer yangbiense) was discovered in 2002 in the self-liu mountain shrubs of villagers in Malutang Village, Shizhong Village, Yangbi County, Cangshan Mountain, Dali Prefecture, Yunnan Province, China. At present, there are only 4 trees left in the wild, and one of them is still felled. Germinated plants, the two larger ones have been able to flower. Acer is recognized as one of the rarest and endangered species in the world, and its conservation and population expansion are the most basic conditions for saving this species. Only when a certain amount of plants are cultivated can researches on conservation biology and environmental biology be carried out, and finally provide material basis, scientific basis and technical support for the s...
