Embodiment 1

[0020] The preparation of embodiment 1 Beauveria bassiana bacterial powder preparation of the present invention

[0021] (1) Cultivation of first-level slant strains:

[0022] 1. Preparation of slant medium:

[0023] Components and proportions of the slant medium: 200g of potatoes, 20g of glucose, 20g of agar, 10g of dried carcasses of peach borer larvae, and 1000mL of water. Its preparation steps:

[0024] ①. Weigh and grind 10g of dried carcass of peach borer larvae, add 1000mL of water and soak for 2-3 hours.

[0025] ②. Peel the potatoes and dig out the buds, weigh 200g, cut into small pieces with a length and width of about 1cm, add them to the soaked dried insect corpses and boil them together, until the potatoes are soft, filter them with gauze, and take the filtrate.

[0026] ③. Add water to the filtrate to 1000mL, add 20g of agar, and boil. Add 20g of glucose after complete dissolution. The medium is at natural pH.

[0027] ④. The filtrate is injected into the t...

Embodiment 2

[0052] The forest application test of embodiment 2 Beauveria bassiana preparation:

[0053] 1. Forest application: In August 2010, the prepared bacterial powder preparation was applied in the jujube orchard of the Fruit Tree Institute of Shanxi Academy of Agricultural Sciences, Taigu County, Shanxi Province. Remove the 1-2cm thick topsoil at the base of the jujube tree with a diameter of about 1 meter, mix the Beauveria bassiana powder preparation with the soil in a ratio of 1:1, spread it evenly, and then cover the topsoil. The dosage is 180g / plant.

[0054] 2. Persistence of Beauveria bassiana in soil

[0055] After the Beauveria bassiana powder preparation was applied in the forest in September 2010, soil samples were taken every two months to detect the activity of Beauveria bassiana spores in the soil, which was measured by the number of colonies and spore production.

Embodiment 3

[0058] The forest application test of embodiment 3 Beauveria bassiana preparation:

[0059] 1. Forest application: In August 2010, the prepared bacterial powder preparation was applied in the jujube garden of Jishan County, Shanxi Province. Remove the 1-2cm thick topsoil at the base of the jujube tree with a diameter of about 1 meter, mix the fungal powder preparation with the soil at a ratio of 1:1, spread it evenly, and then cover the topsoil. The dosage is 180g / plant. At the end of March, 2011, the Beauveria bassiana bacterial powder preparation was applied according to the above-mentioned method, and the consumption was 120g / strain.

[0060] 2. Effect inspection: In the next year, use sampling method to check the control effect, count the healthy larvae and diseased larvae unearthed after overwintering, and the dead worms infected with Beauveria bassiana in overwintering cocoons, and then calculate the corrected mortality rate according to the control. 82.1%-90.5%. In t...

Abstract

The invention provides a beauveria bassiana preparation for controlling peach fruit borers and a preparation method thereof. The preparation method comprises the following steps of: inoculating a strain of beauveria bassiana with the collection number of CGMCC No. 4526 into a test tube loaded with a PDA (potato dextrose agar) slant culture medium containing a healthy peach fruit borer corpse leachate for culture, further performing second-stage strain culture and third-stage solid amplification culture, and finally preparing a bacterial powder preparation. The bacterial powder preparation provided by the invention has a better effect on controlling the peach fruit borers, the control effect can be up to 90.5%, and the fruit-boring rate of an orchard in the same year can be reduced by 83.8% -93.6%. The bacterial powder preparation provided by the invention is particularly suitable for controlling the peach fruit borers in dry and low-temperature regions in the north.

Description

technical field [0001] The invention relates to a microbial preparation, in particular to a Beauveria bassiana preparation for preventing and treating the peach borer and a preparation method thereof. Background technique [0002] Peach borer (abbreviated as peach borer) is a serious pest of fruit tree borers. It generally occurs in northern orchards, endangering the fruits of more than 10 kinds of fruit trees such as apples, jujubes, pears, peaches, apricots, and hawthorns, causing great economic losses to fruit farmers. In the past, chemical insecticides were mainly used to control peach blossoms. Not only will it cause a series of problems such as environmental pollution, killing natural enemy insects, and making pests resistant to pesticides. Moreover, it will cause pesticide residues and reduce the quality of the fruit. The application of entomopathogenic bacteria to carry out biological control is an important way to achieve sustainable control of pests. Since the ...
