Abstract

The present invention is directed to a method of inhibiting aconitase activity of fungal cells in an individual, the method comprising administering an inhibitor of aconitase activity to the fungal cell in an amount effective to inhibit activity of aconitase by said fungal cells.

Description

technical field [0001] The present invention relates to a method of inhibiting the aconitase activity of a fungal cell in an individual, said method comprising administering to said fungal cell an inhibitor of aconitase activity in an amount effective to inhibit the aconitase activity of said fungal cell . Background technique [0002] Without being bound by theory, the antifungal mechanism of action of pyrithione-containing antifungal agents is believed to involve inhibition of aconitase and other mitochondrial iron-sulfur proteins. The mechanism of inhibition is thought to involve the intracellular formation of copper pyrithione by binding to exogenous or endogenous copper and transchelation according to the Irving-Williams series. Copper pyrithione can then penetrate the mitochondrial membrane and interact with iron-sulfur proteins such as aconitase, causing changes in the iron-sulfur active site and resulting inhibition of activity. [0003] In the present invention, Z...
