The preparation is prepared through the following steps: modifying montmorillonite under an acidic condition, mixing the modified montmorillonite, ethyecellulose and chitosan to prepare modified montmorillonite-ethyecellulose-chitosan composite microspheres, and imbedding a papaya seed extract in the microspheres. According to the preparation, edible acid is added into the montmorillonite to modify the montmorillonite, and the modified montmorillonite, ethyecellulose and chitosan are mixed and prepared into the composite sustained-release microspheres, so that the stability of a benzyl isothiocyanate compound in the papaya seed extract imbedded in the microspheres under an acidic condition is guaranteed, and the degradation of the benzyl isothiocyanate compound is restrained. Besides, the papaya seed extract sustained-release preparation can release easily in intestinal juice with a slightly higher pH value, the irritating effect on stomach of benzyl isothiocyanate is reduced, and the bioavailability is greatly improved.
