Embodiment 1

[0039] Example 1: Method for Establishing Rat Inhibitory Lung Cancer Model

[0040] (1) Source of material:

[0041] SD rats: Male SD rats were purchased from the Experimental Animal Center of Anhui Medical University, free of specific pathogens.

[0042] Rat Walker-256 cells: purchased from ATCC.

[0043] (2) Establishment method of rat suppressive lung cancer model

[0044] Step 1: Preparation of Walker-256 cell ascites

[0045]Take out the frozen Walker-256 cells from the liquid nitrogen tank, place them in a water bath at 37°C to dissolve quickly, put them on ice after they are completely melted, draw the cell suspension with a 2ml syringe, and take out the cells that are 3-4 weeks old and weigh 80 -100g male SD rats, hold the rat with their hands, lie supine in their hands, inject into the abdominal cavity from the midline of the abdominal cavity at an angle of 45° between the needle and the abdominal skin, and observe the diet and mental state of the rats every day. ...

Embodiment 2

[0057] Example 2: Inoculation of different concentrations of Walker-256 cells on the impact of lung tumor size

[0058] SD rats were injected with the following mixture:

[0059] 20 μL of mixed solution (10 μL of cell suspension contains 2×10 Walker-256 cells 5 Mix with 10μL Matrigel on ice);

[0060] 15 μL of mixed solution (7.5 μL of cell suspension containing 1.5×10 Walker-256 cells 5 Mix with 7.5μL Matrigel on ice);

[0061] 10 μL of mixed solution (5 μL of cell suspension contains 1.×10 Walker-256 cells 5 Mix with 5μL Matrigel on ice);

[0062] 14 days after inoculation, the rats were killed by dislocation, and the lungs were dissected to observe and measure the size of the foreign bodies in the lungs. The results showed that there were flesh-colored foreign bodies in the left lung of the rats, and the foreign bodies were distributed in the middle of the left lung, and the foreign bodies were of different sizes. , where the maximum diameter of the foreign matter form...

Embodiment 3

[0063] Example 3: After inoculation of the mixed solution containing Walker-256 cells, the detection of tumor growth in rats

[0064] Inoculate 6 SD rats with 10 μL mixture (containing Walker-256 cells 1×10 5 ), 14 days, 21 days, 28 days, 35 days, 42 days after inoculation, the size of the tumor was observed and recorded by CT, and the growth of the lung tumor was analyzed. CT results showed that: 14 days, 21 days, 28 days, 35 days, 42 days after inoculation, the volumes of foreign bodies formed in the lungs of rats were: 0.292±0.042cm 3 , 0.346±0.03cm 3 , 0.388±0.051cm 3 , 0.412±0.038cm 3 , 0.462±0.0571cm 3 . It showed that the inoculated Walker-256 cells grew rapidly in the lungs, which well simulated the invasive growth of lung cancer.

Abstract

The invention relates to a rat transplantable lung cancer model establishing method including the following steps: Walker-256 cell ascitic fluid is prepared; Walker-256 cell is inoculated into SD rat (Sprague-Dawley rat) lung; after anesthesia of a SD rat, left part of thoracic cavity of the SD rat is disinfected by use of povidone-iodine, Walker-256 cell suspension and Matrigel in same volume are mixed evenly on the ice to obtain a mixed solution, the mixed solution is slowly injected between 5th rib and 6th rib at left lung into the left lung by use of an injector, the puncture depth is 0.6-0.8 cm, after the injection, a needle is stopped for 10 s, and then is pulled out slowly, and after the rat is grown for 14 days, a transplantable lung cancer model is established. According to the method, the damage to the rat is small, the Walker-256 cell can be directly transplanted into the left lung only by use of a generally anaesthetized rat, formation of tumor lesion part is clear, the tumor lesion is single and controllable in size, and the method also has the advantages of simple preparation, high tumor formation rate, low animal mortality and the like.

Description

technical field [0001] The invention relates to the field of constructing experimental animal models of diseases, in particular to a method for establishing rat transplanted lung cancer models. Background technique [0002] At present, with the aggravation of air pollution in China, especially the raging smog has brought serious hidden dangers to the health of Chinese people. The International Agency for Research on Cancer under the World Health Organization pointed out that there is sufficient evidence to show that smog is still an important factor in inducing lung cancer. Lung cancer has become one of the cancers leading to the highest mortality rate of cancer patients in the world, and is listed as the focus of future cancer prevention and treatment by the Ministry of Health. However, due to the complex clinical manifestations of lung cancer and the difficulty of early detection and diagnosis, the efficacy of lung cancer cannot be improved. Therefore, replicating an ani...
