Problems solved by technology

After experiments, it was found that this modeling method has the following disadvantages: the operator's operation movements cannot be completely unified, which may easily cause inconsistent stress levels in different rats
After experiments, it was found that this modeling method has the following disadvantages: postoperative rats are prone to excessive bleeding, leading to shock, and shock itself can also induce stress responses in the body, affecting the results of laboratory indicators in rats
After experiments, it was found that this modeling method has the following disadvantages: the area and depth of burns are difficult to grasp, and there are many complications in rats after burns, which can easily induce septic shock and affect subsequent experiments
After experiments, it was found that this modeling method has the following disadvantages: the impact site and force are difficult to grasp, and it is easy to cause rib fractures and lung contusions, and the mortality rate of rats after trauma is high

Embodiment

[0020] A method for modeling rat acute stress response, comprising the following steps:

[0021] (1) Select healthy male SD rats with a body weight of 280g-320g, and randomly divide them into a normal group and a model group. Before the experiment, the rats are fasted for 12 hours and drink water freely;

[0022] (2) Experimental male SD rats are injected with 2% sodium pentobarbital solution at a dose of 4 mg per 100 g of body weight. After SD rats are sedated, they lie on their stomachs, and their limbs are fixed on the animal operating table. Between the S3 and S4 spinal nerves, the tail was cut transversely, that is, the nerve fibers innervating the tail were cut off to simulate the stress response caused by acute peripheral nerve injury.

[0023] (3) Rats in the normal group did not perform any operation.

Abstract

The invention discloses a molding method for acute stress reaction of rats. The molding method comprises the following steps of selecting healthful male SD (sprague dawley) rats with the weight of 280g-320g; injecting 2% of sodium pentobarbital solution into the rats according to the dosage of injecting 4mg of the sodium pentobarbital solution in every 100g of rats; enabling the rats to lie prostrate after the rats are sedative; fixing four limbs of each rat on an animal operation table; transversely cutting off the tail of each rat at the position of a caudalstyle, which is close to a tail end, of the rat, particularly a position between an S3 spinal nerve and an S4 spinal nerve, namely cutting off a nerve fiber for controlling the tail of each rat; and simulating stress response caused by acute injury of peripheral nerves. By the molding method, extra stress response on the rats during molding can be avoided; and the molding method is an optimal mode of producing pure acute stress response. The method does not affect daily activities of the rats, success rate is high, and injury degrees are consistent; and the molding method is easy to operate, stable in result and suitable for standard acute stress response models. Pure, simple and convenient models with quantifiable stress degree can be provided for research on the acute stress response.

Description

technical field [0001] The invention relates to a modeling method of an animal model for medical research, in particular to a modeling method of acute stress response in rats. Background technique [0002] Stress is the body's adaptive comprehensive response to harmful stimuli, and it is the body's adaptive defense mechanism against harmful stimuli. In recent decades, natural disasters such as earthquakes and floods have occurred frequently around the world, and various large-scale gatherings such as the Olympic Games have crowded and stampede incidents, all of which have caused large-scale human trauma, and the types of injuries are mostly trauma caused by acute trauma. The occurrence of acute trauma not only brings severe pain to the injured, but also induces a series of strong stress responses in the body, and may also cause serious complications such as post-traumatic stress disorder due to excessive stress responses. The latest research shows that acute stress can chan...
