Embodiment Construction

[0027] The present invention will be further described below in conjunction with specific examples. The following examples can enable those skilled in the art to understand the present invention more comprehensively, but do not limit the present invention in any way.

[0028] A method for microsatellite genetic marker monitoring of mandarin fish germplasm, comprising the steps of:

[0029] (1) Extraction and detection of genomic DNA

[0030] Genomic DNA was extracted from 0.2 g of caudal fins of 10 mandarin fishes in the same population by phenol-chloroform method. 1% agarose gel electrophoresis was used to detect the integrity of the DNA, and a nucleic acid protein analyzer was used to measure the DNA concentration and purity, and finally diluted with sterile water to about 100ng / μL, and stored at -20°C for later use.

[0031] (2) Search and analysis of microsatellite sequences

[0032] Three mandarin fish of different growth ages in the same population were selected for tr...

Abstract

The invention discloses a method for monitoring Siniperca chuatsi germplasms by virtue of microsatellite genetic marker. The method comprises genome DNA extraction and detection, microsatellite sequence search and analysis, polymorphic microsatellite primer selection and optimization, polymorphic microsatellite detection and monitoring of different geographic siniperca chuatsi populations and hereditary features thereof. Microsatellite loci selection and polymorphic microsatellite marker development and verification are performed on a large number of EST generated by siniperca chuatsi transcriptome sequencing, so that 15 EST-SSR polymorphic microsatellite markers are developed, and different geographic siniperca chuatsi populations and hereditary features thereof can be monitored; the limitations of fuzzy genetic background and less molecular marker amount in existing germplasm detection, selective breeding and germplasm improvement research of siniperca chuatsi can be broken, and a basis is provided for germplasm detection, selective breeding and germplasm improvement of siniperca chuatsi, so that germplasm detection, auxiliary genetic breeding and genomic research processes of siniperca chuatsi molecular markers are promoted.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to a method for monitoring mandarin fish germplasm with microsatellite genetic markers. Background technique [0002] Siniperca chuatsi (Siniperca chuatsi), also known as mandarin fish, osmanthus fish, fat mandarin fish, seasonal flower fish, squid, and flower crucian carp. In biological classification, it belongs to the order Perciformes, the subfamily Siniperidae, and the genus Siniperca genus. It is a rare fish with high economic value. It enjoys the reputation of "freshwater grouper". It has excellent characteristics such as fast growth, tender meat, delicious taste and high nutritional content. It is a famous and special aquatic product loved by consumers and farmers. It is also a nourishing treasure for all ages. However, in recent years, due to the overfishing of rivers, the deterioration of the ecological environment of waters, and the inbreeding in breeding production, etc., ...
