Abstract
The invention discloses a method for molecular typing of capsicum and tomato bacterial scab, and for the first time discloses an operation technique for molecular typing of capsicum and tomato bacterial scab by using a multi-locus sequence typing method (MLST). The invention extracts the genomic DNA of the pathogenic bacteria, screens, amplifies, detects and sequences through seven housekeeping genes. DNAMAN software was used to perform multiple sequence alignment and analysis on the sequencing results to obtain the sequence type (ST) of the strain. Sequence type analysis was performed online using the eBURST V3 program, which can effectively distinguish the four pathogenic bacteria. The 7 housekeeping gene combinations and amplification primers used in this typing technique were adopted for the first time, and the PCR reaction system and conditions were self-optimized. This technical process can be used for taxonomic identification and genetic diversity research of pepper and tomato bacterial scab.
