Abstract
The invention relates to an impedance type electrochemical biosensor based on electrostatic interaction and an application thereof. The impedance type electrochemical biosensor is prepared by the steps of: modifying a gold electrode with SH-DNA; and then modifying the gold electrode with polypeptide containing lysine to prepare the electrochemical biosensor. Polypeptide containing lysine converts between electric neutrality and positive electricity by means of histone acetyltransferase and histone deacetylase, so that the polypeptide is separated from and absorbed on the surface of the electrode to induce impedance change so as to detect the histone acetyltransferase and histone deacetylase. Compared with the prior art, the preparation method of the sensor is simple and low in energy consumption. A linear relationship with the histone acetyltransferase and histone deacetylase is constructed by means of electrochemical impedance change of the electrode, so that two enzymes are detected. Therefore, the sensor has relatively high sensitivity and stability.
Description
technical field The invention belongs to the technical field of biosensors, and in particular relates to an electrostatic-based impedance electrochemical biosensor, a preparation method and an application thereof, which can be used for detection of acetylase and sirtuin. Background technique Protein acetylation, that is, the covalent modification of protein acetyl group, is an important post-translational modification mechanism in the regulation of eukaryotic cells, which can regulate the interaction between proteins in cells, protein stability and signal transduction , in order to regulate physiological responses and disease progression. In the acetylation of different kinds of proteins, lysine acetylation exists as the main post-translational modification in eukaryotes and prokaryotes. Lysine acetylation is a typical "histone code", which is closely related to transcription activity, histone deposition, DNA replication and DNA repair. Histone acetylases transfer the ace...
