Embodiment

[0034] Materials and Methods

[0035] Sequence of IL12p40

[0036] The protein sequence of IL12p40 is shown in NCBI accession number NM_008352 (mouse) or 1F42_A (human), which can be obtained from the website of the National Center for Biotechnology Information (http: / / www.ncbi.nlm.nih.gov / ).

[0037] Cell culture and NSC isolation

[0038] Mouse NSCs were isolated and cultured as in two previous publications (Hsu Y.C. et al., DevelopmentalDynamics2009, 238:302-314; Lee D.C. et al., Molecular and Cellular Neurosciences2009, 41:348-363; these two publications incorporated by reference in its entirety).

[0039] KT98 / F1B-GFP cells were cultured in Dubeca's modified Eagle's medium ( DMEM) / F12(1:1). For NSC isolation, GFP-positive KT98 / F1B-GFP (KT98 / F1B-GFP+) cells were differentiated with a FACSAria cell sorter (BD Bioscience) and cultured in neurosphere formation medium (DMEM / F12, containing 1X B27 ( Gibco), 20ng / ml EGF (PeproTech Inc.), 20ng / ml FGF2 (PeproTech Inc.), 2 μg / ...

Abstract

The present invention provides compositions and methods for enhancing neural regeneration using neural stem cells or at least one component of IL12p40. The composition comprises neural stem cells and neurotrophic factors, which consist of IL12p40 as at least one subunit. The method of enhancing nerve regeneration comprises providing a nerve regeneration composition to an individual, wherein the composition comprises a neurotrophic factor comprising at least IL12p40 as a subunit. The composition of the method may further comprise neural stem cells.

Description

[0001] Cross References to Related Applications [0002] This application claims priority to US Provisional Patent Application No. 62 / 037,612, filed August 15, 2014, the disclosure of which is incorporated herein by reference in its entirety. technical field [0003] The present application relates to compositions and methods for enhancing nerve regeneration using neural stem cells and IL12p40. Background technique [0004] Severe peripheral nerve damage results in reduced activity of motor and sensory neurons and degeneration of nerve fibers and surrounding tissue. Regeneration of damaged peripheral nerves is a complex process, with Wallerian degeneration (WD) as the most basic response, and Schwann cells (Schwann cells) play an important role (Ren Z. et al., Reviews in the Neurosciences 2012, 23:135-143). WD creates a microenvironment for the regeneration of viable neurons and is beneficial for functional recovery (Navarro X. et al., Progress in Neurobiology 2007, 82: 16...
