[0175] Wnt signaling is involved in the development of the hair follicle and serves to initiate the growth phase (anagen phase) of the hair cycle. Using Axin2LacZ / + reporter mice and RNA analysis, it was confirmed that Wnt signaling is elevated during the growth phase (anagen) of the hair cycle. Endogenous Wnt signaling was amplified in the skin and surrounding hair follicles via intradermal injection of R-spondin 2 (Rspo2), and subsequently using Lgr5LacZ / + mice to demonstrate that this enhanced Wnt environment activates Lgr5 in hair follicles - Positive stem cells. Injection by Rspo2 inhibited the onset of the catagen phase (marked by cessation of cell proliferation and increased cell death) and allowed the anagen phase to continue. Therefore, hair growth becomes longer. Rspo2 can be used as a therapeutic protein to target hair follicle stem cells and promote hair growth.
