[0339] Materials and Methods

[0340] Isolation of specific bacterial strains from clothing

[0341] A strain of Brevundimonas sp. isolated from clothing was used in this example. Brevundomonas species were isolated during a study in which bacterial diversity in laundry after washing at 15°C, 40°C and 60°C was investigated. The study was performed on laundry collected from Danish households. For each wash, 20 g of laundry (tea towels, towels, dishcloths, overalls, base t-shirts, t-shirt collars, socks) in the range 4:3:2:2:1:1:1 were used. Washing was performed in a Laundr-O-Meter (LOM) at 15°C, 40°C or 60°C. For washing at 15°C and 40°C, use Ariel Sensitive White & Color laundry detergent (Ariel Sensitive White & Color), and for washing at 60°C, use WFK IEC-A* standard detergent. Ariel Sensitive White and Color Laundry Detergent was prepared by weighing out 5.1 g and adding tap water up to 1000 ml followed by stirring for 5 minutes. WFK IEC-A* standard detergent (avail...

[0351] Preparation of small swatches with biofilm

[0352] Brevundimonas sp. (isolated as described in Example 1) was incubated on Trypticase Soy Agar (TSA) (pH 7.3) (CM0131; Oxoid Ltd., Basingstoke, UK) at 30°C. Pre-grow for 2-5 days. A full circle from one single colony was transferred to 10 mL of TSB and incubated with shaking (240 rpm) at 30°C for 1 day. After propagation, Brevundimonas species were pelleted by centrifugation (Sigma Laboratory Centrifuge 6K15) (3000 g, 7 min at 21° C.) and resuspended in 10 mL of TSB diluted twice with water. Optical density (OD) at 600 nm was measured using a spectrometer (POLARstar Omega (BMG Labtech, Ortenberg, Germany)). Inoculate fresh TSB diluted twice with water to an OD of 0.03 600nm , and 1.6 mL was added to each well of a 12-well polystyrene flat-bottom microplate (3512; Corning Incorporated, Corning, NY, USA) in which no A small round swatch (diameter 2cm) of mycopolyester WFK30A. After incubation at 15° C. for 24 h with ...

Abstract

The present invention concerns a detergent comprising a deoxyribonuclease (DNase). The present invention further relates to methods and uses of the detergent comprising a deoxyribonuclease (DNase) forlaundering.

Description

[0001] References to Sequence Listings [0002] This application contains a Sequence Listing in computer readable form. This computer readable form is hereby incorporated by reference. field of invention [0003] The present invention relates to detergents comprising deoxyribonuclease (DNase). The present invention further relates to methods and uses of detergents comprising deoxyribonuclease (DNase) for laundry washing. Background of the invention [0004] In many natural, industrial, and medical settings, microorganisms typically survive attached to surfaces, encapsulated by extracellular materials including biopolymers and macromolecules. The layer produced by muco-encapsulated microorganisms is called a biofilm. Biofilms are the main mode of bacterial growth in natural environments, and bacteria growing in biofilms exhibit unique physiological properties. Bacteria in biofilms are more resistant to antibiotics, UV irradiation, detergents, and host immune responses tha...
