Abstract

The invention provides a detection method for genotyping of human platelet alloantigen (HPA). The detection method is characterized in that a target fragment is obtained by designing a specific primerand then using a single-tube multiplex PCR amplification reaction; a single base extension primer is designed and then single base extension is performed; then a target DNA sequence is accurately measured by mass spectrometry. The platelet alloantigen HPA-1 to 21 bw allele can be detected at one time. Then a mass spectrum is typed directly, and data are directly output. A whole process is closed,the time is short, and the cost is low. Through detecting the genotyping of the platelet alloantigen of a patient and a blood donor, platelets matching successfully can be found, and the problem of ineffective platelet infusion caused by immune factors is effectively avoided. A detection kit product is low in cost and more accurate, and the detection method is expected to be a routine clinical detection method to replace a serological detection method.
