Embodiment 1

[0040] Pretreatment of pterostilbene ameliorates ConA-induced liver injury in mice:

[0041] Take 20 C57BL healthy male mice of the same age and divide them into 4 groups by random grouping:

[0042] Control group (n=5), using a 1ml syringe, injected DMSO intraperitoneally into the mice, and injected normal saline through the ophthalmic plexus of the mice 1 hour later;

[0043] Con-A group (n=5), using a 1ml syringe, injected DMSO intraperitoneally into the mouse, and injected concanavalin A 10mg / kg through the ocular plexus of the mouse 1h later;

[0044] PTE (trans-3,5-dimethoxy-4-hydroxystilbene) 10mg / kg (n=5), using a 1ml syringe, inject PTE 10mg / kg intraperitoneally into mice, and pass through the eyes of mice 1h later Intravenous plexus injection of concanavalin A10mg / kg;

[0045] PTE 40mg / kg (n=5), use a 1ml syringe, inject PTE 40mg / kg intraperitoneally into mice, and inject concanavalin A 10mg / kg through the mouse ophthalmic plexus 1h later;

[0046] After injecting...

Embodiment 2

[0049] Mechanism of pterostilbene pretreatment improving Cona-induced liver injury in mice:

[0050] Get mouse liver in embodiment 1, extract liver tissue RNA and protein, PTE pretreatment significantly reduces the mRNA level of the inflammatory cytokine INF-γ and TNF-α induced by ConA ( Figure 5 ). Compared with the ConA group, the expression of INF-γ and TNF-α decreased by 2.7 times and 3.1 times (in the PTE 10mg / kg group, p<0.0001) and 3.3 times and 4.5 times (in the PTE 40mg / kg group, p<0.0001) respectively. 0.0001). Given that the hypercoagulable state in the liver aggravates fibrin deposition and accelerates extensive liver necrosis.

[0051] The role of PTE in the intrahepatic hypercoagulable state was investigated. Since TF and Fibrin factors are highly expressed in the hypercoagulable state, the role of PTE in the intrahepatic hypercoagulable state was observed by detecting the expression of these two factors. see results Figure 6 , TF (approximately 43%) was s...

Abstract

The invention provides application of pterostilbene to preparation of drugs for preventing acute liver injuries, and belongs to the technical field of disease prevention. The pterostilbene is trans-3-,5-dimethoxy-4-hydroxystyrene. According to the application, the pterostilbene reduces expression of an inflammation cytokine INF-gamma and an inflammation cytonf-kine TNF-alpha, the fibrous protein content in a liver is decreased, phosphorylation of MAPK and NF-kappa B is inhibited, macrophage accumulation is reduced, tissue factor production is inhibited, and the level of glutamic oxalacetic transaminase and glutamic-pyruvic transaminase is decreased to prevent the acute liver injuries.

Description

technical field [0001] The invention belongs to the technical field of disease prevention, and in particular relates to the application of pterostilbene in the preparation of medicines for preventing acute liver injury. Background technique [0002] In the past few years, due to factors such as drug abuse and viral infection, acute liver injury has been increasing worldwide, threatening people's lives and health. Acute liver injury can lead to the release of inflammatory cytokines, oxidative stress overload, hepatocyte apoptosis, and elevated alanine aminotransferase (ALT) and aspartate aminotransferase (AST). At present, drugs such as glucocorticoids and azathioprine can be used to alleviate liver damage, but the serious side effects of drugs will affect the quality of life of patients. Therefore, there is an urgent need to determine and develop an effective treatment strategy for liver injury Vaccination recommendations, J Am Acad Dermatol., 2017, 76(2), 191-198.]. [0...
