Embodiment 1

[0036] Example 1: Panobinostat's effect on improving the behavior of mice with experimental autoimmune encephalomyelitis

[0037] 1.1 Establishment and administration of experimental autoimmune encephalomyelitis mouse model

[0038] Experimental principle

[0039] MOG for C57BL / 6 mice 35-55 Induce experimental autoimmune encephalomyelitis (experimentallyallergic encephalomyelitis, EAE) model.

[0040] experimental method

[0041] Female C57BL / 6 mice, 6-8 weeks old, weighing 18-20g, were subcutaneously injected with MOG after 1 week of adaptive feeding 35- 55 300 μg and 600 ng of pertussis toxin were injected intraperitoneally to establish an animal model of experimental autoimmune encephalomyelitis (EAE). A normal control group was set, n=10. After 11 days of modeling, the animals began to develop symptoms, with varying degrees of limb paralysis. Mice with limb paralysis were selected as the experimental autoimmune encephalomyelitis model.

Embodiment 2

[0084] Example 2: Effect of panobinostat on inflammatory cell infiltration and demyelination in the spinal cord of EAE model mice

[0085] 2.1 Establishment of EAE mouse model and drug administration

[0086]Experimental principles, experimental methods, and experimental results are the same as in Example 1.1.

[0087] 2.2 Effect of panobinostat on the infiltration of inflammatory cells in the spinal cord of EAE mice

[0088] experimental method

[0089] On the 28th day after modeling, 3 mice in each group were anesthetized with 4% chloral hydrate. Perfuse with normal saline first, wait until the color of the liver turns white, and then perfuse with 4% paraformaldehyde until the limbs of the animal become stiff. The head was decapitated, and the lumbar enlarged part of the spinal cord was fixed in 4% paraformaldehyde. Paraffin sections were made from the lumbar spinal cord and stained with hematoxylin-eosin (H&E) to observe the infiltration of inflammatory cells in the spi...

Abstract

The invention discloses application of panobinostat in preparation of a medicine for preventing, relieving and/or treating multiple sclerosis. In a mouse experimental autoimmune encephalomyelitis model, panobinostat shows a good treatment effect, and can effectively treat pathological changes and progression of diseases. The therapeutic effect is achieved by inhibiting proliferation of microglial cells in a central nervous system.

Description

technical field [0001] The invention relates to the application of panobinostat in the preparation of drugs for preventing, alleviating and / or treating multiple sclerosis, and belongs to the technical field of medicine. Background technique [0002] Multiple sclerosis (MS) is an autoimmune disease characterized by inflammatory demyelinating lesions of the central nervous system, which has the characteristics of high recurrence and disability. Clinical manifestations include paralysis, numbness, cramps, aphasia, and visual impairment. Epidemiological studies have shown that 50-300 people in every 100,000 people suffer from multiple sclerosis, and there are 2.3 million people in the world suffering from multiple sclerosis, mainly concentrated in European and American countries. The prevalence of multiple sclerosis increases with latitude, and the incidence of multiple sclerosis in women is twice that of men (Neurology.2014; 83(11):1022-1024). The etiology of multiple scleros...
